ObjectiveThrough the comparison of the Expression of MMP-9, TIMP-1in carotidatherosclerotic plaque and Normal arterial tissues, and the expression of MMP-9, TIMP-1,Hcy in patients with carotid atherosclerosis and normal control, to explore the relationshipbetween these factors and the stability of plaque and the role they played in thedestabilization process of carotid atherosclerotic plaque.MethodsTake20carotid atherosclerotic plaques which were removed from patients withcarotid artery stenosis who were performed carotid endarterectomy in our hospital as thestudy group, divided into2groups(stable plaques and unstable plaques)according topreoperative carotid artery ultrasonography. Take15Normal arterial tissues which wereremoved from patients without atherosclerosis who were performed surgery in ourhospital as the normal control. All of the specimens were excised in4%paraformaldehydefor12h, conventional dehydration, paraffin embedding, HE staining to observe themorphological differences, of MMP-9and TIMP-1in the plaques. All the patients weredrewed venous blood, for detect serum HCY, MMP-9, TIMP-1by elisa in the morning ofthe day taken operation.Results1. With the development of atherosclerotic disease, serum concentrations of MMP-9,TIMP-1, Hcy tended to increase; the serum Hcy, MMP-9, TIMP-1of experimental groupconcentration was significantly higher than that of normal control group (p <0.05);serumHcy, MMP-9, TIMP-1concentration of unstable plaque group is higher than the stableplaque group (p <0.05).2. The expression of MMP-9、TIMP-1in atherosclerotic plaque and normal arterywere significant differences (p <0.05), and was significantly higher than the stable plaquegroup in the expression of unstable plaque group (p <0.05). 3. The serum MMP-9/TIMP-1ratio in the experimental group、SP group、and UPgroup were significant differences (p <0.05).4. In the normal control group, SP group, UP group, Hcy was positively correlatedwith MMP-9(p <0.01).5. In the normal control group, the serum Hcy is highly correlated with TIMP-1(r=0.842p=0.000), there was no correlation between the serum levels of Hcy and TIMP-1in group SP (r=0.668p=0.070), there was no correlation between the serum levels of Hcyand TIMP-1in group UP (r=-0.14p=0.657).6. In the normal control group, serum Hcy concentration and MMP-9/TIMP-1ratiohas no correlation (r=0.054p=0.849). In SP group, serum Hcy concentration andMMP-9/TIMP-1ratio does not exist (r=0.344p=0.404). The UP concentration of serumHcy and MMP-9/TIMP-1ratio were highly correlated (r=0.746p=0.005).Conclusions1. Serum Hcy, MMP-9, TIMP-1level can reflect the severity of atheroscleroticdisease in a certain extent.2. The ratio of serum MMP-9/TIMP-1can reflect the stability of atheroscleroticplaque.3. Along with the increase of serum HCY level, serum,and atherosclerotic plaqueMMP-9increased. The result is that MMP-9/TIMP-1imbalance, accelerate the progress ofplaque instability.
