Purpose: Traditional Chinese features and modern preparation techniques arecombined in this topic, extraction, concentration, drying process,preparation and quality standard of the drug are under systemic research.Meanwhile,anti-virus test of the obtained preparations has been conducted toensure the safety and therapeutic.Methods:EC50in the anti-virus test has been the index of screening extractionsolvent in this article. The method of orthogonal trial has been applied tooptimize the extraction condition of forsythia, honeysuckle and arctium, withthe index of chlorogenic acid, forsythiaside, arctigenin and extractsrate.The aqueous extract of forsythia, honeysuckle has been concentrated,dried and pulverized with some accessory for the demand of dosage forms. Andthe condition of dry granulating was optimized with the index of yield ofgranules. HPLC, TLC method has been conducted on the quality control toinstitute reasonable quality standard. Also,the toxic effect action on cellsand the inhibition of respiratory syncytial virus caused by preparation havebeen investigated.Results:The extraction solvent was50%ethanol and EC50was339μg·mL-1;Theoptimum extraction condition:6fold50%ethanol marinated3times at90℃, and1h each time, the optimal extraction rate of chlorogenic acid, forsythiaside, arctigenin and extracts rate were2.68%,0,32%,7.98%and29.4%;The extract was concentrated with decompression at60℃to be creamwhose relative density was1.08-1.10(55℃), and dried the cream in vacuumdrying chamber at60℃,-0.085MPa, crushed it into fine powder when themoisture content was less than5%,finally mixed it with0.8%stevioside and2%talcum powder until evenly.The optimum granulation condition of drygranulator machine: frequency conversion of feeding was20.0-21.0Hz,frequency conversion of tabletting was25.0-26.0Hz, frequency conversion ofgranulating was25.0-26.0Hz,the pressure of the roller was2MPa. Thegranulation rate can reach53.64%, and the number will be up to90%whengranulated3times. The angleofrepose of the granule was32.8°,the bulkdensity was0.7771g·mL-1,and CRH was57%.Qualitative and quantitative analysisfor preparation composition of three different batches were conducted withTLC and HPLC and we established the thin layer identification method anddetermined the limit of standard substances.Preparations on respiratorysyncytial virus by the antivirus test proved that the TC50was2.3937mg·mL-1,theEC50was52.03μg·mL-1,and the index of treatment was46.01.Conclusion: The stable extraction technology optimized in this topic makesit able to enrich the active constituent, the taste of preparations can beameliorated when mixed with stevioside, the lubricity can be increased whilethe dilutability can be reduced when mixed with talcum powder, and thegranulating technology is shown to stable with high yield of granules. Simplemethod of quality standards, sharp spectrum of a graph and goodreproducibility contribute to the quality of the preparations.Thepreparations obtained show a good inhibition of respiratory syncytial virusby the antivirus test.
