| Objective: In order to provide reference for Radix Aconiti Lateralis praeparataprocessing and establish quality control to evaluate different varieties of Radix Aconiti Lateralis praeparata quality,we performed experiments of extracting and identifying chemical composition of Radix Aconiti Lateralis praeparata,establishing fingerprint for different varieties of Heishunpian,exploring the alkaloids distribution in Radix Aconiti Lateralis praeparata, and analizing the reasonable processing methods and discussing the quality of Radix Aconiti Lateralis praeparata effectively.Methods: Materia Medica Research was carried out including,name, plant form and types, origin and production environment, functions and main therapy,processingmethods, tropism of taste, breed and medicinal parts research. By silica gel column chromatography, Sephadex LH-20column chromatogramphy and other modern means of separation, the chemical composition of the root of Radix Aconiti Lateralis praeparata. were separated and purified,combined with the physical and chemical properties and MS,1H-NMR,13C-NMR and other modern spectroscopic methods, identifing structures of the compounds.Using high performance liquid chromatography (HPLC) method, the fingerprint of Heishunpian was established, alone with the content of aconitine, mesaconitine and hypaconitine in different transversal and longitudinal parts, were determined respectively.Results: The herbal research shows that, the name recorded in the herbal, mainly of Jin, Gen, Gen, Jian,Ji,Geng,San Jian,San Jin and so on. The genuine producing area of Radix Aconite Lateralis is Sichuan(Jiangyouã€Pingwuã€Mianyangã€Anxian and Butuo) and Shanxi(Zhengguã€Erxianã€Mianxianã€Nanzhengã€Hanzhong).The quality of Radix Aconite Lateralis produced in Sichuan and Shanxi is better than other’s. The best harvest time and processing technology in herbal record are co nsistent to those ofmodern,record.Chemical composition of Radix Aconiti Lateralis preparata and the content of bioactive components in different varieties and different methods-processingdrugs.It can be easily drawed the conclusion that,the content of alkaloids in Heishunpian is higher than that in Baifupian. It can be known that10peaks are calibrated of the fingerprint and the similarity is higher than85%.Four compounds includingbeta sitosterol,palmitic acid,2,3-2hydroxy propyl palmitate and mesaconitine wereisolated and identified from chloroform extraction of the root of Radix Aconiti Lateralis praeparata.Conclusion: The distribution of aconitine, mesaconitine and hypaconitine in Radix Aconiti Lateralis praeparata obeys certain rules, which can provide reference for Radix Aconiti Lateralis praeparata processing and quality control. Combined withcontent determination, the specificity of fingerprint can be used to control the drug’quality. Besides, it can also provide the basis for further control processing procedure. |
