Objective: This study aimed to verify whether direct myocardialinjection of BMSCs could improve cardiac function, progress ventricularremodeling and ameliorate myocardial perfusion, and also provideexperimental basis for the treatment of myocardial infarction with bonemarrow mesenchymal stem cells transplantation.Methods:20fisher rats were chosen to construct acute myocardialinfarction model through LAD ligation.16fisher rats survived, and99mTc-MIBI gated SPECT was applied to detect myocardial perfusion(uptake rate of99mTc-MIBI and the number of under-perfusionsegment)and cardiac function including ejection fraction, LV end-systolicvolume, LV end-diastolic volume. Then the16fisher rats were randomlydivided into two groups, the control group and the experiment group. Inthe control group, after one week of LAD ligation, the saline was directlyinjected into myocardium; in the experimental group, after one week ofLAD ligation, the BMSCs was injected directly into the myocardium inthe number of2x106.99mTc-MIBI gated SPECT was applied to detect thescope of the myocardial infarction, myocardial perfusion and cardiac function in the experimental group and the control group. At last all of thehearts were removed for histological examination.Results: After treatment, in the saline injection control group, the EFvalue has the downward trend, but no significant difference betweenpre-injection and post-injection (P>0.05); in the experimental group, fourweeks later after BMSCs treatment, EF was significantly increasedcompared with pre-treatment with significant difference(F=26.51,P=0.024). In the control group, before and after treatment, the LVEDVand LVESV showed a trend towards dilatation, and there were significantdifferences(F=25.09,P=0.031,F=21.27,P=0.040); in the BMSCstreatment group, after treatment, the LVESV reduced, but has nostatistical difference (P>0.05). The LVEDV increased but without anystatistical difference(P>0.05). Before and after follow-up,in the controlgroup, myocardial perfusion showed that the number of infarct segmentsdeclined, the99mTc-MIBI uptake continued to decline, but there was nosignificant difference (P>0.05),While in the experimental group thenumber of infarct segments declined(F=29.15,P=0.011), and the uptakerate of99mTc-MIBI in segmental infarction increased(F=37.22,P=0.009).Histological examination also confirmed that BMSCs treatment coulddecrease infarct size and increase ventricular wall thickness.Conclusion: BMSCs treatment of acute myocardial infarction in micecan improve cardiac function and enhance the value of EF, and can improve myocardial reperfusion and decrease the infarct areas, but couldnot progress left ventricular remodeling.
