| Objective: To explore the optimal harvesting and growing periodand establish HPLC fingerprint on the different harvesting period of FructusGardeniae in Bazhong;Methods: HPLC method was used to determine thecontents of geniposide, crocinâ… , and ursolic acid. The data were analyzed byadopting t-test and Z score multi-index test,and the best harvesting andgrowing period with ideal contents were choosed. The content determination ofursolic acid was carried out on ELSD with drift tube was heated to60℃and aXDB C18(4.6mm×150mm,5μm) heated to25℃inthe columncompartment. The mobile phase was acetonitrile-water (81:19) at a flow rateof1mL·min-1.HPLC fingerprints were obtained on a Kromasil C18(9.2mm×250mm,5μm)column with mobile phase of acetonitrile-waterat. Theflow rate of1mL·min-1,to detect signal with variable wavelength in wholeprocess and the column temperature was maintained at35℃. Results: Results oft-test showed that content of crocinâ… was increasedfrom green fruit(non-maturity) to red fruit(maturity) of30-year-old Fructus Gardeniae;content ofgeniposide was decreased from green fruit to red fruit;content of ursolic acidwas the highest in green fruit while it was the lowest in yellow fruit;thecontents of geniposide, crocinâ… , ursolic acid of30-year-old red fruit weresignificantly higher than those of3-year-old one. After comparison andanalysis,the fingerprints were variant on the different harvesting period of Fructus Gardeniae in Bazhong, the Similarity was reduced from greenfruit(non-maturity) to red fruit(maturity) of non-maturity. Conclusion: It isof great significance to optimal the harvesting and growing period byinvestigation of geniposide, crocinâ… , and ursolic acidand to establish HPLCfingerprint on the different harvesting period which not only can ensure thequality of Fructus Gardenia from Bazhong, but also will provide an reliablescientific basis for the follow-up research. |
