| Objective: To examine the expression of ERC1(excision repaircross-complementing1,ERCC1) and BRCA1(breast cancer susceptibilitygene1) in nasopharyngeal carcinoma(NPC)tissues and its relationship withchemosensitivity to cisplatin-based chemotherapy. Methods: To collectionEighty-four advanced nasopharyngeal carcinoma diagnosed untreatedpatients,who were diagnosed at Oncology,Affiliated Hospital of LuzhouMedical College from February1,2012to February28,2013, were treated withPF regimen (cisplatin80mg/m2first day+5-FU1000mg/m2continuousintravenous drip×4days, repeated every4weeks×2cycles)2cycles ofinduction chemotherapy. To Completed two cycles of induction chemotherapybefore concurrent chemoradiotherapy. By immunohistochemistry to detectERCC1and BRCA1expression in84patients with nasopharyngeal carcinoma.Last statistical analysis of nasopharyngeal carcinoma of ERCC1and BRCA1expression and the relationship of the efficacy of chemotherapy.Results:Expression of ERCC1was found mainly in the nucleus of NPC, few incytoplasm. Expression of BRCA1was also found in the nucleus of NPC. Thepositive rate of ERCC1expression and BRCA1in NPC were47.62%and52.38%respectively. Expression of ERCC1and BRCA1in NPC was unrelated with patients’ gender, age, ECOG score, histological type,histological differentiation, chemotherapy, TNM stage (P>0.05). Highexpression of ERCC1group of patients with nasopharyngeal carcinomaeffective rate was77.50%(31/40), and low expression group effective rate was95.45%(42/44), the difference between the two was statistically significant (P=0.015). High expression of BRCA1group of patients with nasopharyngealcarcinoma effective rate was79.55%(35/44), and low expression groupeffective rate was95.0%(38/40), the difference between the two was statisticallysignificant (P=0.036). The expression of ERCC1and BRCA1expression inNPC was positively correlated (r=0.553, P<0.05). Conclusions: Theexpression of ERCC1and BRCA1in nasopharyngeal carcinoma wereunrelated with its clinical features. The expression of ERCC1and BRCA1inNPC was negatively correlated with platinum-based chemotherapy. Theexpression of ERCC1and BRCA1at the protein level in nasopharyngealcarcinoma was positively correlated. |
