ObjectiveTo investigate the expression of ERCC1 gene in gastric cancer cell lines and influence on cisplatin resistance of gastric cancer cells.MethodsSGC7901/DDP, SGC7901 cell lines were cultured routinely with Complete RPMI-1640medium. and stable expression of mRNA and protein of ERCC1 were detected by RT-PCR and Western Blot. The SGC7901 cell lines were treated with 5.0μg/ml concentrations of cisplatin for 0,24,48,72,96h,and then treated with 0,1.25,2.5,5.0,10.0μg/ ml concentrations of Cisplatin 48h respectively.The expression of ERCC1 was observed by RT-PCR and Western Blot.Finally, MTT were performed to detected cell survival rate and calculated median inhibitory concentration (IC50) and resistance index after treatment of cisplatin.Results1.ERCC1 gene expression existed in SGC7901/DDP, SGC7901, BGC-803 cell lines, The results of RT-PCR reaction and Western Blot test suggest that resistant cell line SGC7901/DDP mRNA and protein levels of ERCC1gene were significantly higher than the other two groups, (P <0.05). Treated with 5μg/ml cisplatin for 0,24,48,72,96h,mRNA and protein was detected in SGC7901 cells,the results showed that the expression of ERCC1 was up-regulated with time extension. SGC7901 cells were exposed for 48 h to different concentrations of cisplatin. When treated with low concentration of cisplatin , ERCC1expression demonstrated increased in SGC7901.with the increase of the concentration of cisplatin, ERCC1 gene displayed decline status of expression. Cisplatin IC50 values of SGC7901/DDP,SGC7901 cell were 15.70±0.37μg/ml, 5.53±0.13μg/ml respectively. MTT assay showed that expression of ERCC1 gene in gastric cancer cells was related with IC50 index of cisplatin, the quantity of ERCC1 expression in drug-resistant cells was significantly higher than that of sensitive cells.Conclusion1.The expression of ERCC1 gene is related with cisplatin-resistance of gastric cancer.2 Cisplatin can induce increased expression of ERCC1,this increase was time- and concentration-dependent.
