Objective: To determine the expression of heat shock protein27(HSP27)and Glypican-3(GPC3)in primary hepatocellular carcinoma(PHC) and benign liver disease to investigate the relationship between theirexpression and that the diagnosis and clinical pathological features of PHC。Methods: Immunohistochemistry was used to detect the expression ofHSP27and GPC3in32cases of primary hepatocellular carcinoma tissues,30cases of cirrhosis of the liver tissues,25cases of benign liver placeholdertissues,31cases of hepatitis tissues. The relationship between theirexpression and clinical factors of PHC was analyzed.Results: HSP27was mainly expressed in the cytoplasm of liver cancercells and minority expressed in cell membrane. HSP27was highly expressedin PHC tissues (positive rate of56.25%), while it was not expressed in theadjacent tissues and benign liver tissues. The difference was statisticallysignificant (p<0.05). The expression of HSP27was related with PHC gradeand TNM stage,but it was no significant correlation with the patient’s age,gender, tumor size, HBsAg characterization and serum AFP values.GPC3 was mainly expressed in the cytoplasm of liver cancer cells. GPC3washighly expressed in PHC tissues (positive rate of87.5%), while it was notexpressed in the adjacent tissues and benign liver tissues. the difference wassignificant (p<0.05). The expression of GPC3was not significant correlationwith patient’s clinical and pathological features of PHC。Conclusion: HSP27and GPC3were all available for the differentialdiagnosis of primary liver cancer and benign liver tissues. GPC3have moresignificant sensitivity in primary liver cancer, it may become a newpathological diagnosis marker of liver cancer. The expression of HSP27in alower degree differentiation and metastasis cases of tumor was higher, whichmay be more valuable in the severity and prognosis of liver cancer patients.
