Backgroud and Objective:MicroRNAs(miRNA) was a class of small noncoding RNAs,which contains20~25nucleotides,it can regulate differentiation,proliferation and apoptosis of cells either by affecting the stability of thetarget mRNA or by inhibiting its translation and transcription. Atpresent,the research data demonstrated that miRNAs plays an importantrole in the development of some skin diseases,but the research on atopicdermatitis(AD)-related miRNAs was absent.In this article,we firstscreened differentially expressed miRNAs in plasma of AD comparedwith normal controle by using the miRCURY LNATMchip; Then weverify the accuracy of the gene chip results by RT-qPCR and Looking forspecific miRNAs in plasma of AD.We hoped that miRNA as a newbiomarker of disease diagnosis,the result of this study can providetheoretical support and experimental data of AD.Further research mayplay a helpful role for the pathogenesis,clinical diagnosis and theclinical evaluation of treatment of AD.Materials and methods:5ml peripheral venous blood were collected from9patients with ADand6normal persons,under the condition of free-enzyme,we extracted the total RNA from the15samples.We screened and analysised thedifferentially expression of miRNAs by using the miRCURY LNATMgene chip (Exiqon Corporation).miRNAs gene expression profile of ADwas preliminarily Established. Then some significant miRNAs ofup-regulated or down-regulated expression were furtherly verified byRT-qPCR and analysed.Results:1. The number of up-regulated miRNAs genes In plasma of AD was105and the number of down-regulated miRNAs genes was225.2. In the up-regulated miRNAs gene,over2-fold chang was61,over3-fold chang and less than5-fold chang was17,over5-fold chang was5.At the same time, the down-regulated miRNAs gene,less than0.5-foldchang was170(P<0.05).Conclusions:1. The expression profile miRNAs in plasma of AD was preliminarilyestablished.Compared with normal healthy control,61up-regulatedmiRNAs and170down-regulated miRNAs in AD were screened.2.Through the literature according to the experiment,differentiallyexpressed miRNA in AD were selected for subsequent RT-qPCRverification,The miRNA genes were miR-142-5p、miR-1237-3p andmiR-4275、miR-5047.
