Objective To investigate the effects and the possible mechanism ofinhaling budesonid in treating bleomycin-induced pulmonary fibrosis inrats.Methods Forty-five SD rats were randomly divided into threegroups(15in each group):a control group,a model group and a interventiongroup.Rats in the model group and intervention group were given a singledose of intratracheal bleomycin,and the rats in control group were receivedequal volume of0.9%normal saline to bleomycin.The intervention groupinhaled budesonid every two days after7days intratracheal instillation,whilethe model group and control group inhaled the same volume of0.9%normalsaline.Five rats in each group were randomly killed on the7th,the14th andthe28th day after atomizing inhalation and make a collection of specimensrespectively,HE and Masson staining was used to determine the extent ofalveolus inflammation and pulmonary fibrosis;Immunohistochemicaltechnique was used to evaluate the level of transforming growthfactor(TGF-β) and connective tissue growth factor(CTGF) in lung tissues;The serum and BALF samples were harvested for Elisa Test for TGF-β and CTGF;RT-PCR was used to learn the level of typeⅠand Ⅲ collagen in lungtissues.Results HE and Masson staining of lung tissues approved succeedingin copying the classical model of pulmonary fibrosis, The extent of alveolusinflammation and pulmonary fibrosis in the model group and interveninggroup was obviously more serious than that in the control group(p<0.05).Besides, the intervening group was better than that in the modelgroup(p<0.05).Compared with the control group,significant over-expressionof CTGF and TGF-β was shown in lung tissue of the model group and theintervention group(P<0.05),the latter was lower than that in the modelgroup(P<0.05); The result of Elisa Test showed the level of CTGF andTGF-β in BALF and serum of each group was different,indeed,theexpression of CTGF and TGF-β in the BALF was significantly higher thanthat in the serum. The expression of CTGF and TGF-β in the model groupand intervening group was higher than that in the control group(p<0.05),theintervening group was less than that in the model group(p<0.05);TypeⅠandⅢ collagen in the model group was significantly higher than that in thecontrol group(P<0.05),but the intervention group was less than the modelgroup (P<0.05).Conclusion Budesonid exerts anti-fibrosis potency by reducing the production of type I and Ⅲ collangen through inhibition of cytokines suchas TGF-β and CTGF in tissues and serum....
