Purpose: Using cell culture in vitro technology, RT-PCR(Reverse transcriptasepolymerase chain reaction) and Western blot technique, Observing the reaction of ILK, CTGFprotein and mRNA. Researching the effect of CTGF-ILK signaling pathway on human lungfibroblasts, and the intervention by Xiankekeli on pulmonary fibroblast.Method:1.Observing the expression of CTGF,ILK mRNA and the protein inHLF-02,which was cultured in vitro, with the RT-PCR and Western-blot Method.2.Observing the expression of ILK mRNA and Protein,and detecting the α-SMA, FN,I-Collagen, III-Collagen, with the RT-PCR and Western-bolt Method, after HLF-02wasstimulated by CTGF in different concentration(5ng/ml,15ng/ml,25ng/ml,50ng/ml).3. AfterHLF-02was stimulused by CTGF, add Xiankekeli to it, then detected the expression of ILK,α-SMA, FN, I-Collagen, III-Collagen, mRNA and protein. Observing the effect on humanlung fibroblasts of Xiankekeli from low to high dose.Result:1, Human lung fibroblasts cells could express CTGF, ILK mRNA and protein.2,Stimulated by CTGF in different concentration,the trend of human lung fibroblasts mRNAand protein expression was the same, and the expression of ILK, FN, I-collagen, III-collagen was increaded, and it had a positive correlation with the concentration.3, Xiankekelicould inhibit the proliferation of human lung fibroblasts, reduce the expression of ILK, I-collagen, III-collagen, FN protein. Compared with the blank group, the effect of high dosegroup was obvious, it had significant differences (p <0.05); Compared with prednisone group,high dose group had not significant differences (p>0.05).Conclusion:1. Under the stimulation of CTFG with different concentration, theexpresstion of ILK,FN, I-Collagen, III-Collagen mRNA and protein increaded had a positivecorrelation with the concentration. It meand that CTGF-ILK signaling pathways existed inpulmonary fibrosis. FN, I-Collagen, III-Collagen may be the key signaling factor.2.Xiankekeli had the effect of against pulmonary fibrosis. through the inhibition ofhuman lung fibroblasts proliferation, reducing the expression of ILK, FN, I-collagen and III-collagen mRNA and protein. Xiankekeli may play a role in antifibrosis by blocking or partial inhibition of CTGF-ILK signal pathway. This may be one of the mechanisms of Xiankekeli inthe treatment of pulmonary fibrosis.3.Xiankekeli can reduce the expression of FN, I-collagen, III-collagen mRNA andprotein, with the most significant effect in high dose group. Xiankekeli may play a role inantifibrosis by restraining the expression FN, Collagen I and Collagen III and decreasing theextracellular matrix. This may be one of the mechanisms of Xiankekeli in the treatment ofpulmonary fibrosis.
