Objective:To observe VM in differently differentiated gastric cancercells in vitro, and to explore the possible underlying mechanisms bytransfecting SGC7901cells with Mig-7-siRNA and observing the impacton VM formation.Methods:①The capability of VM formation in differentlydifferentiated gastric cancer cells were observed by light microscopy andscanning electron microscopy in three-dimensional culture system;②Theexpression of Mig-7were detected by RT-PCR;③The impact oftransfection with Mig-7-siRNA into SGC7901cells on the abilities of VMformation, invasion and migration were examined;④The expression ofMig-7, p-ERK1/2and MMP-2in SGC7901cells were analyzedrespectively by western blot.Results:①VM formation was observed in poorly differentiatedMKN45and moderately differentiated SGC7901cells, but not in welldifferentiated MKN28and normal GES-1cells in vitro;②Mig-7wasexpressed in MKN45and SGC7901cells, but not in MKN28and GES-1cells;③The abilities of VM formation, invasion and migration were changed by transfection with Mig-7-siRNA into SGC7901cells.;④Theformation of VM was reduced by down-regulating the expression ofp-ERK1/2and MMP-2protein in SGC7901cells.Conclusion: Mig-7is expressed in gastric cancer cells which are ableto form VM. The formation of VM is inhibited by reducing the expressionof Mig-7in gastric cancer cells, which might be mediated bydown-regulating the expression of p-ERK1/2and MMP-2protein.
