BACKGROUD & OBJECTIVE: Vasculogenic mimicry (VM),a microcirculation structure formed by malignant tumor cells simulating endothelial cells,is a new microcirculation form of tumor cells which found resent years.There are few resaerches about vasculogenic mimicry in gastric cancer till now.This research observed whether gastric cancer SGC-7901 cells can form vasculogenic mimicry in vitro 3-dimentional culture and had a primer examination on its mechanism.METHODS: Put SGC-7901 cells in vitro 3-dimentinal culture and observed if it could form vasculogenic mimicry.Immunohischemistry tested the MMP-2 and MT1-MMP expression of 3-dimentional cultured SGC-7901 cells.RT-PCR,Western Blot tested the effect of PI3K way inhibitor LY294002 on MT1-MMP mRNA and MMP-2 protein level respectively.Observe the effection of LY294002 on SGC-7901 cells vasculogenic mimicry quantity.RESULTS: Gastric cancer SGC-7901 cells could form vasculogenic mimicry in vitro 3-demintional culture. MT1-MMP and MMP-2 highly expressed in gastric cancer cells that took part in the tube structure.MT1-MMPmRNA,MMP-2 protein level and vasculogenic mimicry quantity of teated group decreased obviously comparing with that of control proup(p<0.05 all).CONCLUSION: Gastric cancer SGC-7901 cells had the ability to form vasculogenic mimicry in vitro 3-dimentional culture.MT1-MMP and MMP-2 took part in SGC-7901 cells vasculogenic mimicry.Activiation of PI3K signaling pathway could promote its happening.One of the mechanism was probably through increasing MT1-MMP and MMP-2 expressions.
