Background: Cardiovascular and Cerebrovascular diseases aregrievous threats to human health, and the morbidity are rising along with theaging of the population. Atherosclerosis is the pathological basis ofCardiovascular and Cerebrovascular diseases.Therefore, to prevent theoccurrence and development of the AS is a key step to prevention andtreatment of Cardiovascular and Cerebrovascular diseases. Antiplateletagents and statins are already widely used in clinical treatment, which hadbeen proved to be classical and efficient.Because of deeper understandingof atherosclerosis mechanisms, antiatherosclerotic drugs are still keepprogressing. As antioxidant could inhibit atherosclerosis, so recent studiespay more attention to this effect.Probucal, which is a representative drug ofAntioxidant, could inhibit atherosclerosis, attributing to regulating bloodlipids,anti-oxidation,anti-inflammation and improving endothelial function。But the study about effect of Probucol on anti-atherosclerosis is lessreported.Objceive: To investigate the effect of probucol, aspirin, andatovastatin combination treatment on anti-atherosclerosis. Method:20patients with carotid atherosclerosis were randomlyand double-blind divided into the PAS treatment group and AS controlgroup.10patients in PAS group were treated withprobucol(0.375g,twice/d),aspirin(100mg/d),and atorvastatin(10mg/d).10patients in AS control group were treated with aspirin(100mg/d),andatorvastatin(10mg/d). All patients received treatments were followed up for12months. The serum lipids(TC,TG,LDL-C,HDL-C,ox-LDL),High-sensitivity c-reactive protein (hs-CRP), carotid intimal medialthickness(IMT),and the value of the longest line diameter multiple themaximum thickness of carotid plaque were measured before and after12months of treatment. Measurements and comparisons were performedbefore and after treatment in each group.Results:The levels of TC,LDL-C,ox-LDL in patients’ serum in bothgroups were reduced (P<0.05)after treatment,especially in PAS group(P<0.05).The carotid IMT and the value of the longest line diametermultiple the maximum thickness of carotid plaque were reduced in bothgroups after treatment (P<0.05),especially in PAS group (P<0.05).Conclusions: Combintion treatments of probucol, aspirin andatorvastatin reduces the levels of TC, LDL-C, and ox-LDL in serum,andboth of carotid IMT and the size of carotid plaque more prominent,compared to control group. Our data demonstrates PAS treatment was moreefficient on anti-atherosclerosis. This study demonstrates Probucol can strengthen the effect of aspirin and atorvastatin on anti-atherosclerosis.
