Objective:(1) To observe the expression of Hesl at different developmental stages inthe SD rat spinal cord.(2) To observe the expression of Hes1at different time afterspinal cord injury in adult SD rats.(3) To observe which cell types that Hes1to expressby immunofluorescence staining technique.Methods:(1) To create the SD rat spinalcord tissue at different developmental stages: E15.5d (embryonic15.5days), E18.5d(embryonic18.5days), P0d (birth), P3d (born3days), P7d (born7days), P2w (born2weeks), P4w (born4weeks), Adult (2) To establish to the adult rat spinal cord injurymodel, and to take spinal cord tissue after injury6h,12h,1d,2d,3d,5d,7d.(3) Todetect the expression of Hes1at different stages of development and different stagesafter spinal cord injury in spinal cord by RT-PCR (4) To detect the expression of Hes1at different developmental stages and different stages after spinal cord injury in ratspinal by western blot (5) Using immunofluorescence techniques to observe theexpression of Hes1in normal development at different stages in the rat spinalcord.While using Hesl\GFAP, Hesl\MBP, Hesl\β-Tubulin, Hesl\Nestin double labelingstaining to detect the positive cells which express Hes1in different stages ofdevelopment.Results:(1)The expression of Hes1is in different developmental stages inrats spinal cord.In the development of rat spinal cord(E15.5,E18.5,P0,P3,P7,P2W),Hes1gene expression and protein levels were significantly higher than in adulthood (P<0.01).(2) The expression of Hes1in3-5days after injury was higher in the mRNAlevels and protein levels(P<0.01).(3) Immunofluorescence staining results showed thatGFAP-positive cells in E15.5d of the rat spinal cord are not observed, and that in P0, P7,P2w spinal cord appears; MBP-positive cells were not observed in the embryonic, but inP7, P2W; β-Tubulin-positive cells can be detected in E15.5, P0, P7, P2W; Nestinpositive cells can be observed in the embryonic, and that in P7, P2W testednegative.Conclusion:Hesl were expressed in various developmental stages of SD ratspinal cord, In developmental stages hes1has a higher expression in spinal cord of SDrats.(2) Hes1has a higher expression after spinal cord injury.(3) The differentiationand development of nerve cells in rats showed Temporal and spatial expressionlaw.(4)Hes1expressed in NSCs, mature neurons, oligodendrocytes and astrocytes.
