| Objective:(1)To establish the tissue specimen library of SD rat spinal cord from embryo to adult.(2)To observe the expression and cell location of transcription factor Oligl in rat spinal cord.(3)To investigate the law of Oligl expression in SD rat spinal cord from embryo to adult.Methods:The rat spinal cords at embryonic days14.5(E14.5d),18.5(E18.5d), postnatal (P) days P-0d, P-3d, P-7d, P-2w, P-4w and adult were collected to established the tissue specimen library of SD rat spinal cord from embryo to adult.(1) To extract messenger RNA (mRNA) and detect the expression of oligl mRNA using the reverse transcription polymerase chain reaction (RT-PCR).(2) To detect the change of Oligl expression at mRNA and protein levels in the rat spinal cord at different developmental periods using Real-Time PCR and Western Blotting, respectively.(3)To detect the expression and cell location of Oligl in different neural cells of rat spinal cord at different periods using immunofluorescence staining technique.Results:(1) RT-PCR, Western blotting and immunofluorescence staining showed that Oligl could be detected in the spinal cords of SD rats from E14.5d to adult.(2) The double-label immunofluorescence staining showed that the oligl expressed in both04-positive cells and beta-Tublin-positive cells, but not in MBP-positive cells and GFAP-positive cells in the spinal cords of P7d and P2w.(3) The Real-Time PCR and Western blotting results showed that the expressions of oligl mRNA and protein were detected in the developing rat spinal cord from E14.5d to adult. However, the expression levels of Oligl at all developmental periods didn’t keep a same level and didn’t gradually increase or decrease with the process of development. The expression of Oligl was higher in the early embryos, but decreased gradually to the lowest level at P7d, and then increased gradually again.Conclusion:(1) The transcription factors Oligl can be detected in the spinal cord of SD rats from E14.5d to adult.(2) The expression of Oligl is higher in the early embryos, but decrease gradually to the lowest level at P7d, and then increase gradually again.(3) The Oligl mainly expresses in immature oligodendrocyte precursors cells and neurons, but not in mature oligodendrocytes and astrocytes. |
