Lamivudine is one of the pyrimidine nucleoside antiviral drugs, mainly be used forthe treatment of chronic hepatitis. Valsartan is an orally active specific angiotensin Ⅱreceptor antagonist, for the treatment of mild to moderate essential hypertension. Tocontrol the content of invalid enantiomer in lamivudine and valsartan as well as tocontrapose the shortcomings of the existing chiral separation methods. The experimentused capillary zone electrophoresis for chiral separation at the first time. The ultimatelyestablished methods could be used to detect the samples of lamivudine and valsartan.The capillary zone electrophoresis method for chiral separation of lamivudine: it wasseparated with an untreated fused silica capillary (75μm I.D., effective length was55cm).40mmol/L sodium dihydrogen phosphate and40mg/mL HP-β-CD (pH=3.10) was runningbuffer solution at the detection wavelength of270nm. Separation voltage was15kv andthe injection time of sample was15s. Under this condition, the separation of lamivudineand its enantiomer could be achieved1.60, greater than1.5which is required in theChinese pharmacopoeia. Moreover,the method of methodology verification was standard.And the method was available for the detection of the sample of lamivudine. At the sametime, it was compared with the pharmacopeia method.The capillary zone electrophoresis method for chiral separation of valsartan:complete separation of (R,S)-enantiomers of valsartan with a very good resolution(Rs=1.64) was achieved within an untreated fused silica capillary (75μm I.D., effectivelength was60cm).30mmol/L sodium acetate and18mg/mL β-CD (pH=4.50) wasrunning buffer solution at the detection wavelength of254nm. Separation voltage was20kv and the injection time of sample was5s. In addition, the methodology verification ofthis method was fully met. And it could be used for valsartan testing. The method and theHPLC in pharmacopeia were compared.
