| High altitude polycythemia (HAPC) is a common and frequently encountered disease in plateau area in China,threatens people’s health who living and working in plateau area Seriously. Traditional Tibetan medicine as the World recognized medical science has the unique advantage of treating HAPC. The San Guo Tang San must take one before Performing bloodletting therapy, putting surgery into effect and Disease blood flow out to the body to the treatment purpose. But Tibetan bloodletting and San Guo Tang San of HAPC intervention mechanisms have not been reported.ObjectiveUnder the guidance of traditional Tibetan medicine theory, using metabonomics methods, observe mechanisms of San Guo Tang San powder control the animal model of HAPC caused by chronic hypoxia and the patient of HAPC, To study the effect of drug effect and mechanism of action.Method(1) In the5000m Plateau environment simulation cabin anoxic40days (22h/d), SD rats for the animal model of HAPC.To observe effectiveness and Mechanisms.(2) Using chloroformic acid ethyl ester derivative method and GC-MS hyphenated techniques chromatography, investigate the change of Endogenous small molecule metabolites and Prevention of protection of San Guo Tang San decoction. By chemometrics method principal component analysis and partial least squares discriminant analysis,the Small molecule endogenous metabolites has changed obviously.(3) Collecting blood from the patient in Lhasa area, all of them are up to standard of HAPC, Male Hb≥210g/L, Women Hb>190g/L, and checking up blood routine examination, hemorheology.(4) As the research object to patients, using chloroformic acid ethyl ester derivative plasma method of GC-MS, by chemometrics method principal component analysis and partial least squares discriminant analysis,study the blood of the patient’s blood before and after treatment. Looking for new methods of treating HAPC.Result(1) Researches show that San Guo Tang San can prevent HAPC, there are differences between Indicators.Metabonomics study found there are differences between blank group San Guo Tang San decoction high-dose, San Guo Tang San decoction low-dose and rhodiola group,and found endogenous substance.There are9-Hexylheptadecane Glycine, N-methyl-N-methoxycarbonyl-, ethyl ester2-chloro-Octane,2,4-Di-tert-butylphenol.(2) According to clinical statistic,there are visible differences between before and after bleeding, and Symptoms alleviated, researches show that Medication therapy and blood-letting therapy metabolic profiling all tend to separate. With statistical methods chould observe the difference of plasma Metabolic profiling.to explain the scientific basis of combinen San Guo Tang San with Occupational Therapy. And provide the basis for the treatment of HAPC.ConclusionThere is significantly different metabolic profiling between various groups of HAPC rats and HAPC patients through treatment. By PCA and PLS-DA to determine the endogenous metabolic profiles of plasma metabolites. Through the blood of the indicators observed San Guo Tang San scattered and bloodletting on the plateau polycythemia have some improvement. |
