High altitude polycythemia(HAPC)is a disease characterized by compensatory polycythemia or complications caused by chronic high altitude hypoxic environment,threatening the health of high-altitude residents.The mechanism is complex,and there is no clear theory.At present,the diagnosis of HAPC is single and mainly depends on subjective intention.Micro RNA(miRNA)is a class of non-coding single-stranded small RNAs that have received high attention and great application value in recent years,and their research as diagnostic markers is becoming more and more mature.In this study,the specific expression of miRNAs in HAPC was carried out.The specific contents are as follows:Objective:To obtain the miRNA expression profile in peripheral plasma of HAPC patients,and to combine with bioinformatics analysis to explore the pathogenesis of HAPC and to provide reliable markers for its early diagnosis.Methods:The peripheral plasma miRNAs of HAPC patients and healthy controls at the same altitude were detected,and the miRNA expression profiles in the plasma of HAPC patients were comprehensively analyzed from the genome level,and the specifically expressed miRNAs were screened.The peripheral plasma samples of20 HAPC patients with Hb≤21g/dl but with obvious clinical symptoms and 18 healthy controls at the same altitude were collected,and q RT-PCR was performed to verify the specific expression of miRNA.Results:The expression profiles of the obtained miRNAs were analyzed,and there are 8 differentially expressed genes were obtained: hsa-miR-486-5p,hsa-miR-451 a,hsa-let-7b-5p,hsa-miR-486-3p,hsa-miR-144-5p,hsa-miR-183-5p,hsa-miR-194-5p,hsa-miR-1180-3p,and they were all up-regulated in HAPC,and in the q RT-PCR verification,we found hsa-miR-486-5p,hsa-miR-451 a,hsa-miR-486-3p,hsa-miR-194-5p gradually increased with the progression of the disease.Conclusion:hsa-miR-486-5p,hsa-miR-451 a,hsa-miR-486-3p,hsa-miR-194-5p may be involved in occurrence and development of HAPC,they may be used as a molecular marker for the early diagnosis of HAPC.
