Background Esophageal cancer is the eighth most common cancer worldwide, with a wide variation in its frequency between high-and low-incidence regions. In our country, Linzhou and Taihang mountains is high-incidence regions. Esophageal epithelial carcinogenesis involve multiple genes changed, eventually lead to the cell cycle regulation out of control and malignant proliferation of cells.E26transformation-specific-1(also known as Ets-1) gene is a proto-oncogene and has homology with the v-ets-1retro viral gene sequence which was originally i solated from the avian leukemia retrovirus E26. The protein encoded by Ets-1is a transcription factor, which mediates specific DNA binding to the core sequence, the n regulates gene transcription, participates in extracellular matrix degradation, cell m igration, tumor angiogenesis, and other links.Signal transducers and activators of transcription6(also known as STAT6) is one of seven members of the STAT protein family, which is composed of847amino acids. It participates in the signal transduction of IL-4and IL-13. It is the key regulator of T cell growth, differentiation and Thl/Th2balance and plays an important role in B cell proliferation differentiation and the production of IgE and the same type conversion. It related with the bronchial asthma and allergic disease in the clinical. Recent studies found that STAT6signaling pathway can be activated in the part of the tumors, such as Hodgkin’s lymphoma, breast cancer can be activated, but whether it play a role in esophageal cancer still need to be further elaborated.Objective To explore expression of E26transformation-specific-1and Signal transducers and activators of transcription6in esophageal lesions and to analyze its correlation with clinicopathologic characteristics. Methods The expression levels of Ets-1and STAT6protein and mRNA were detected in36cases of esophageal squamous cell carcinoma(ESCC),36cases of dysplasia and18cases of normal esophageal tissues by immunohistochemistry St reptavidin-peroxidase (S-P、RT-PCR and Western Blotting.Results1. Positive expression rates of Ets-1and STAT6proteins were mainly localized in cytoplasm or nucleus of ESCC. There were low or no expressions in normal esophageal tissues. Positive expression rates of Ets-1and STAT6proteins were gradually increased from normal esophageal tissues, atypical hyperplasia to ESCC tissues, there were significantly difference among the groups (,P<0.05).2. The relative amount of Ets-1and STAT6mRNAs were gradually increased from normal esophageal tissues, atypical hyperplasia to ESCC tissues, there were significantly difference among the groups (.P<0.05).3. The relative amount of Ets-1and STAT6proteins were gradually increased from normal esophageal tissues, atypical hyperplasia to ESCC tissues, there were significantly difference among the groups (P<0.05).4. The abnormality expression of Ets-1and STAT6were positively correlated with tumor grade, infiltration and lymphatic metastasis of ESCC tissue.5. Expressions of Ets-1and STAT6proteins appeared positive correlation of ESCC (r=0.447, P<0.05).Conclusions1. The abnormal expression of Ets-1and STAT6were related to cancerization of squamous epithelium of esophagus and they may have synergistic effect.2. Ets-1and STAT6are possible to be the molecular indicators to judge the malignant biological behaviour of the patients with esophageal cancer.
