Objective:To observe the Chinese medicine Bugan decoction on the Achilles tendon and sciatic nerve of rats with experimental diabetes insulin-like growth factor-1(IGF-1) expression of the intervention effect.Methods:36male SD rats were randomly divided into normal control group, model-control group and Bugan decoction-treated group.1%of streptozotocin (STZ)-induced diabetic rats, blood glucose≥16.7mmol/1for successful modeling. The rats of the treatment group were administered with Bugan decoction(28.6g/kg.d)by intragastric injection for4weeks,while the model group administered with the same dose of saline. Observe about changes of hair, behavior, weight and blood glucose dynamically. Rats were killed after measured weight and tested fasting blood glucose in4th week.IGF-1peptide content of Achilles tendon by enzyme-linked Immunosorbent assay(ELISA). Immunohis-tochemical method and ELISA were used to test the level of protein of IGF-1in sciatic nerve.Results:(1) Bugan decoction has no significant ameliorating to wei ght and blood glucose of the rats with diabetes induced by streptoz otocin (P>0.05)。(2) The effects of BG decoction on the IGF-1pr otein and peptide content in sciatic nerve:results of model group a nd treatment group were both lower than that of normal control gro up(P<0.01),the treatment group was better than the model group, th e results were statistically significant(P<0.05).(3) The effects of BG decoction on the IGF-1peptide content in Achilles tendon:rat Ac hilles tendon IGF-1of model group and treatment group were high er than those in the normal group(P<0.05);there is no significant dif ference (P>0.05)between the treatment group and the model group.Conclusions(1)In high glucose conditions, expressions of IGF-1protein, IG F-1peptide contents in sciatic nerve both decreased and weakened t he repair capacity of nerve fibers, which may be one of the reason of damages to structure and function of peripheral nerves.(2)Bugan decoction can increase the expressions of IGF-1prote in and IGF-1peptide contents in sciatic nerve; it could proect the s ciatic nerve certain extent via provoking the protection mechanism of IGF-1.(3)In high glucose conditions, the IGF-1peptide contents in Ac hilles tendon increased, and Bugan decoction has no effects to this. The negative results could be concerned with untimely detection ti me point of the model and insufficient monitoring point of the sam ple.
