Objective To compare the distribution and morphological features of Aβ immunoreactive products in the hippocampus, cortex and amygdaloid nucleus of the APP/PSl double transgenic mouse (2xTg) and APP/PSl/Tau triple transgenic mouse (3xTg) models of Alzheimer’s disease.Method12-months-old2xTg and the age-matched mouse of3xTg were used in this experiment.6E10immunohistochemical staining was used to show the distribution and morphological features of A(3immunoreactive products, and quantitative analysised the difference of the products by an image analysis system.Result The deposition of Aβ in the hippocampus, cortex and amygdaloid nucleus of2xTg were mainly seen as extracellular Aβ(eAP), and formed a lot of positive plaques; the positive products were distributed as intracellularly Aβ(iAβ) for the3xTg, but the Aβ positive plaques were rarely seen in the three regions.Conclusion The differences of Aβ positive products distribution between the2xTg and3xTg is probably to be related to the differences of neuropathological changes of these two AD models.
