The technique of immunohistochemistry and in situ hybridization were used to detect the expressions of OPN, CD44v6, MMP-9in laryngeal squamous cell carcinoma and normal laryngeal mucosa tissues, thus to investigate the correlation between clinical pathological features and the expressions which OPN, CD44v6, MMP-9mRNA and their protein had in laryngeal squamous cell carcinoma and normal laryngeal mucosa tissues; To investigate the relationships among OPN, CD44v6, MMP-9mRNA and their protein in laryngeal squamous cell carcinoma.The protein and mRNA expressions of OPN, CD44v6, and MMP-9were detected in47cases of laryngeal squamous cell carcinoma tissues, and10cases of normal laryngeal mucosa tissues through the technique of immunohistochemistry (IHC) and in situ hybridization (ISH) respectively. The correlation between the three indexes and such clinical and pathological features of patients as age, sex, type, the size of tumor, the degree of tumor differentiation, TNM stages and lymph node metastasis, was analyzed, the correlation among the three indexes was analyzed, and the consistence of expression of OPN, CD44v6, MMP-9was detected between the two techniques of immunohistochemistry (IHC) and in situ hybridization (ISH).As the protein expressions of OPN, CD44v6, MMP-9was detected in47cases of the laryngeal squamous cell carcinoma tissues and10cases of normal laryngeal mucosa tissues through the technique of IHC, the positive expression rates of OPN protein, CD44v6protein and MMP-9protein were63.8%(30/47),76.6%(36/47) and68.1%(32/47) respectively in laryngeal squamous cell carcinoma tissues, and apparently higher than10%(1/10),30%(3/10) and0%(0/10) in normal laryngeal mucosa tissues, the variance was significant (P<0.05); and the high expression of OPN and CD44v6protein in laryngeal squamous cell carcinoma tissues had correlation with the degree of tumor differentiation, TNM stages and lymph node metastasis, however, it didn’t have relationship with the clinical and pathological features such as age, sex and the location of tumor. The high expression of MMP-9protein in laryngeal squamous cell carcinoma tissues had correlation with TNM stages and lymph node metastasis, but didn’t have relationship with patients’ age, sex, the location of tumor and the degree of tumor differentiation.Of47tissue specimens of laryngeal squamous cell carcinoma, the positive expression of OPN protein were30cases, CD44v6protein were36cases, and MMP-9protein were32cases. of30cases with positive expression of OPN protein, the positive expression of CD44v6protein were27cases and MMP-9were24cases. As the result of Spearman correlation analysis, the expression of OPN protein and CD44v6protein was significantly positive, the expression of OPN protein and MMP-9protein was significantly positive, and the expression of CD44v6protein and MMP-9protein was significantly positive as well.As the expressions of OPN mRNA, CD44v6mRNA, MMP-9mRNA were detected in47cases of the laryngeal squamous cell carcinoma tissues and10cases of normal laryngeal mucosa tissues through the technique of ISH, the positive expression rates of OPN mRNA, CD44v6mRNA and MMP-9mRNA were61.7%(29/47),72.3%(34/47) and66%(31/47) respectively in laryngeal squamous cell carcinoma tissues, and apparently higher than10%(1/10),30%(3/10) and0%(0/10) in normal laryngeal mucosa tissues, the variance was significant (P<0.05); and the high expression of OPN mRNA and CD44v6mRNA in laryngeal squamous cell carcinoma tissues had correlation with the degree of tumor differentiation, TNM stages and lymph node metastasis, however didn’t have relationship with such clinical and pathological features as age,sex and the location. The high expression of MMP-9mRNA in laryngeal squamous cell carcinoma tissues had correlation with TNM stages and lymph node metastasis, but didn’t have relationship with patients’age, sex, the location of tumor and the degree of tumor differentiation.Of47tissue specimens of laryngeal squamous cell carcinoma, the positive expression of OPN mRNA were29cases, CD44v6mRNA were34cases, and MMP-9mRNA were31cases. of29cases with positive expression of OPN mRNA, the positive expression of CD44v6mRNA were26cases and MMP-9were24cases. As the result of Spearman correlation analysis, the expression of OPN mRNA and CD44v6mRNA was significantly positive, the expression of OPN mRNA and MMP-9mRNA was significantly positive, and the expression of CD44v6mRNA and MMP-9mRNA was significantly positive as well.Of47tissue specimens of laryngeal squamous cell carcinoma, the positive expression of OPN protein were30cases, of30cases, the positive experession of OPN mRNA were29cases; the positive expression of CD44v6protein were36cases, of36cases, the positive expression of CD44v6mRNA were34cases; the positive expression of MMP-9protein were32cases, out of32cases, the positive expression of MMP-9mRNA were31cases, through Kappa testing analysis, the value of P is smaller than0.01, that further proved the two technologies utilized for research in this project had a very high consistency.The expressions of OPN, CD44v6, MMP-9in laryngeal squamous cell carcinoma tissues were apparently higher than that in normal laryngeal mucosa tissues, which pointed out that the high expression of OPN, CD44v6and MMP-9could possibly be related to the development of laryngeal squamous cell carcinoma; High consistency was found in detecting OPN, CD44v6, MMP-9between immunohistochemisty and in situ hybridization; As concluded by IHC and ISH that the expressions of OPN, CD44v6, MMP-9in cases with lymph node metastasis were apparently higher than the positive expression of that in cases without lymph node metastasis, the high expression of OPN, CD44v6, MMP-9was probably related to metastasis of esophageal carcinoma; that the correlation between the expressions of OPN, CD44v6, MMP-9and the range of tumor infiltration was positive, OPN’s positive expression was related to the invasion of esophageal carcinoma; As pointed out by the rank correlation analysis, the expression of CD44v6and MMP-9was apparently correlated with OPN, which indicated that the synergistic action of OPN, CD44v6and MMP-9was apparently noted in the metastasis and invasion of laryngeal squamous cell carcinoma, and the increase of OPN expression could possibly cause the increase of CD44v6and MMP-9expressions, thus caused the metastasis and invasion of tumor. In a conclusion, the codetection of OPN, CD44v6and MMP-9might be very important and meaningful to foresee early-stage of metastasis and prognosis of tumor and OPN could be a potential therapeutic target in laryngeal squamous cell carcinoma.
