Background:cerculer smooth musle around the bile duct and pancreatic duct at theend of the circular smooth muscle and pancreatic biliary ampulla called as Oddisphincter.Dysfunction of Oddi sphinter(SOD) gradually known: its not only leading linkmost biliary, pancreatic disease pathogenesis, but also is the key of the secondarypathological change of these disorders. To explore the pathogenesis of SOD and find theeffective treatment method is an important subject in the field of digestive disease.Traditional Chinese medicine thinks, SOD is a hypochondriac pain, jaundice. Peony andlicorice decoction used to relieve pain, disharmony of liver and spleen, sour-sweet herbsnourishing Yin, with spasmolytic and relaxation of smooth muscle function, also can beODDI sphincter, reduce the pressure, but the exact mechanism is unclear. Researchshows: high cholesterol can cause rabbit SOD, associated with increased [Ca2+]i andcalcium channel change. In this study, using the method of ELISA, sphincter musclering contraction function determination and other test means, with normal andhypercholesterolemic rabbits ODDI sphincter muscle and muscle cell as the researchobject, research of peony and licorice decoction serum and its effective components ofpaeoniflorin in normal and hypercholesterolemic rabbit sphincter of ODDI ring, ring ofmuscle cells and muscle movement, three inositol phosphate (IP3),[Ca2+]i mechanism,the effect of calcium, determine the signal transduction mechanism of its regulation.Study of paeoniflorin on gastrointestinal smooth muscle relaxation process,decrease the concentration of [Ca2+]i can induce relaxation of gastrointestinal smoothmuscle. Whether the vasodilation effect of the same and the same signal transductionmechanism of the sphincter of ODDI?Preliminary observation of paeoniflorin in Hypercholesterolemia Rabbits in vitro effects of ODDI sphincter muscle ring contraction activity, observed in vitro,paeoniflorin with diastolic function on hypercholesterolemia of sphincter of ODDI.Further studies are required to determine its specific mechanism. This subject is afurther research, in hypercholesterolemic rabbits as the model of SOD, the peony andlicorice decoction serum and its effective component of paeoniflorin as the researchobject, to study its effect on normal and hypercholesterolemic rabbit sphincter of ODDIby ELISA method to measure the concentration of rabbit IP3means, signal transductionmechanism from the angle of cell information and cell ion channels into their role incells, the regulation of ODDI sphincter cells (normal and dysfunction) dynamicmechanism, so as to investigate the peony liquorice decoction in the treatment of SODand provide a theoretical basis to a certain extent.Objective:1Effect of Shaoyaogancao Decoction serum on isolated hypercholesterolemiarabbit sphincter of ODDI muscle contractile activity2peony and licorice decoction, paeoniflorin in rabbits of normal and highcholesterol three inositol phosphate (IP3) concentrationMethods1) establish high cholesterol induced ODDI sphincter dysfunction modelAccording to reports in the literature (Wei Jingguo):16pure New Zealand rabbitswere randomly divided into two groups, the control group (control, eoN) andhypercholesterolemia (hypereholesterolemia, HC) model group with8rats in eachgroup, the control group was fed with standard diet, the experimental group was fedwith standard diet and cholesterol, were fed for eight weeks, induced by highcholesterol in rabbits.2) ODDIsphincter muscle ring making methodMuscle ring end by stainless steel wire hook is fixed on the bottom of the muscleZao hook on the other end of the hook and the muscular tension transducer.3) of pure New Zealand rabbits24, divided into8groups,3rats in each group,peony and licorice decoction orally,2times/day, a total of three days, after the lastadministration, respectively, to5points,15points,30points,45points,60points,90points,120points,180points8time points after femoral artery blood, instrument tomeasure the concentration of paeoniflorin and glycyrrhizic acid in serum by highperformance liquid chromatography, curve drawing blood drug concentration, themaximum plasma concentration time of serum samples containing blood sampling time. 4) Hypercholesterolemia Rabbits6,9normal rabbits, rabbit SO, divided into fivegroups, with three rats in each group were measured by ELISA, IP3concentration.Results:1hypercholesterolemia group SO intracellular IP3concentration was higher thanthat of the normal;2peony and licorice decoction drug-containing serum group inhypercholesterolemic rabbit SO muscle relaxant strength higher than the control group,and with the increase of serum concentration, diastolic amplitude increase;3peony and licorice decoction serum, paeoniflorin group IP3concentration wassignificantly lower than the control group;Conclusion:1high cholesterol can cause increased IP3concentration in rabbit SO cells;Theconcentration of2peony and licorice decoction medicine-containing serum can reducehypercholesterolemia rabbit SO cell IP33The change of IP3concentration directly affect the SO contractile activity.
