| Purpose: To observe jiedutongluobaoshen capsule’s effect to diabeticrats’ podocyte ZO-1, and to observe the interventional effect to the diabeticnephropathy rats’ and the rats’ podoctye ZO-1by the help of animal model, andto find a new way of prevention and cure for diabetic nephropathy.Method:80Wistar rats were regularly fed, free access to water for5days,and they were randomly divided into16groups by weight,5rats for each group.2groups were randomly extracted as normal group which were normally fed. Another14groups were feed with high fat diet for5weeks modeling. The successfulmodeling rats were randomly divided by weight into3groups which is Chinesemedicine experimental group, western medicine control group and blank group.The Chinese medicine experimental group was administrated with gavage ofjiedutongluobaoshen capsule, western medicine control group was administratedwith gavage of Rosiglitazone, blank group was administrated with gavage of thesame volume of normal saline. After12weeks administration we collect thespecimen and to compare the specimen. Automatic biochemical instrument was usedto do the Biochemical tests. HE staining was made and to observe renalmorphological changes. And to detect ZO-1express by immunohistochemical method.Results: By the animal model experiment, we found that jiedutongluobaoshencapsule can improve the experimental rats general state, blood glucose, bloodlipids, renal function and it also can reduce the rats’ urine output andurinary albumin, and we also observed the ZO-1standard’s rise.Conclusion: Jiedutongluobaoshen capsule has a protect effect to rats’kidney, and the protection mechanisms maybe related to DM rats’ renal ZO-1. |
