| Respiratory Syncytial Virus (Respiratory Syncytial Viral, RSV) which causes infants with lower respiratory tract infection is the most important viral pathogen in the world-wide in the recent years, which is impact with severe clinical symptoms and high mortality, so it attracts high attention.RSV can lead to imbalance of T helper cells Th1/Th2after being infected, induce the expression of related cytokines, causing the body’s immune damage. RSV is the important ligands of toll-like receptors (Toll-like receptors), which can induce the activation of TLRs signaling transduction pathways, causing the secretion of downstream cytokines, such as IL-6and IL-8and IL-1beta. Jin Xin oral liquid developed by Mentor professor Wang shouchuan is confirmed with resistance to RSV infection and immune regulation in previous clinical and experimental study, its effective monomer resveratrol also has antiviral effects. The mechanism of antagonizing RSV infection is not yet clear, so this experiment starts with toll-like receptor signal transduction pathway induced by RSV infection, investigates the regulatory role of Jinxin oral liquid and resveratrol on the expression of inflammatory factor, and researches the correlation to other TLRs pathways, speculates that the molecular immunology mechanism of antagonizing RSV infection.Objective1To observe the expression levels of IL-6, IL-8and IL-1β infected by RSV both in vitro models and vivo models, so as to explore the possible mechanism in the aspect of immunology.2To observe the regulatory role of Jin Xin oral liquid and resveratrol on the expression of IL-6, IL-8and IL-1β infected by RSV both in vitro models and vivo models, so as to explore the immunological mechanism of antagonizing RSV infection..MethodsThis experiment adopts the experimental zoology, cell culture, molecular immunology and ELISA method to detect the expression of IL-1β in alveolar lavage of BALB/c mice and the expression of IL-6and IL-8in hep-2cells supernatants. The statistical soft wares of EXCEL and SPSS17.0were used to analyze the experimental data.Results Experiments in vivo:24h after RSV infecting BALB/c mice, the expression of IL-1β significantly increased (P<0.01), the expression of IL-1β in both Jin Xin high prevention group and equivalent prevention group decreased, and that in the equivalent prevention group decreased significantly (P<0.01);72h after RSV infecting BALB/c mice, the expression of IL-1β significantly increased (P<0.05), but it decreased slightly compared with that at24h. Jin Xin oral liquid down-regulate IL-1β expression. Both Jinxin high prevention group and treatment group down-regulated the expression of IL-1β function compared with RSV group, but there was no statistically significant difference between them(P>0.05).72h after RSV infecting BALB/c mice, the expression of IL-1β significantly increased compared with that at24h and72h (P<0.05), but it increased compared with the normal group. Both Jin Xin treatment groups can down-regulate the expression. Experiments in vitro:24h after RAW264.7cells infected by RSV, in cell supernatants the expression of IL-1β the supernatant is very low.24h after hep-2cells infected by RSV, the expressions of IL-6and IL-8in RSV group increased significantly compared with the control group (P<0.01), and they were down-regulated in JinXin oral liquid groups compared with RSV group (P>0.05, P<0.01) OResveratrol in the dosage of50μmol/L can typically down-regulate the expression of IL-6(P<0.01) and IL-8(P<0.05), which effect is the same with the dosage of25μmol/L, but there was no statistically significant (P>0.05).Conclutions1. Jin Xin oral liquid significantly alleviate the lung and body’s inflammation of BALB/c mice infected by RSV.2. The expression of IL-6, IL-8and IL-1β are down-regulated both in vitro models and vivo models with RSV infection by Jin Xin oral liquid and Resveratrol.
