A Research Of The Potential Correlation Between Th17Cells、Its Related Cytokines And The Development Multiple Myeloma

Objective：To detect the levels of Th17cells and its related cytokines in peripheral blood ofmultiple myeloma patients，try to analyze the probable potential correlation betweenTh17cells and the development multiple myeloma。Methods：Selected39MM patients who diagnosed and treated in the second affiliatedhospital of nanchang university from September2011to June2012, mark them asexperimental group； and selected30healthy controls who was having healthexamination， they didn’t infected by helicobacter pylori、 hepatitis B virus、cytomegalovirus、 HIV、 syphilisand didn’t suffer from autoimmune disease、malignancy，mark them as control group。Collected the peripheral blood of patientsmarked as experimental group and control group.1、Flow cytometry was used toanalyze the frequency of Th17cells in CD4+T cells in MM pantients and healthycontrols。2、RT-PCR was used to adopted to detect the expression of RORγt mRNA inMM pantients and healthy controls which is the specific transcription factor of Th17cells。3、ELISA was employed to measure the sernm density of IL-17in MMpantients and healthy controls which is the specific secretory production of Th17cells。4、Then analyze the relationship between the levels of Th17cells and theclinical characteristics of MM patients，include gender、age、ISS stage、type、globulin、light chain、M-protein、plasmocyte in bone marrow、β2-microglobulin andefficacy。Results：1、The frequency of Th17cells in MM patients is3.509±0.594%，the frequencyof Th17cells in health controls is2.913±0.846%。The frequency of Th17cells in MMpatients is significant higher than those in normal controls（P<0.05）。2、The expression of RORγt mRNA in MM pantients is0.568±0.177，theexpression of RORγt mRNA in health controls is0.468±0.163。The expression of RORγt mRNA in MM pantients is significant higher than those in normal controls（P<0.05）。3、The density of IL-17in MM pantients is27.200±4.666pg/mL，the density ofIL-17in health controls is23.758±7.410pg/mL。The density of IL-17in MMpantients is significant higher than those in normal controls（P<0.05）。4、The frequency of Th17cells shows positive correlation with ISS stage、β2-microglobulin and efficacy of MM patients（P<0.05）；while it showed littlecorrelation with gender、age、type、D-S stage、globulin、light chain、M-protein、plasmocyte of MM patients（P>0.05）。Conclusion：In this research，we detected the levels of Th17cells and its related cytokines inperipheral blood of multiple myeloma patients，and analyze the probable potentialcorrelation between Th17cells and the development multiple myeloma。The resultindicate that the frequency of Th17cells、the expression of RORγt mRNA which isthe specific transcription factor of Th17cells、the density of IL-17which is thespecific secretory production of Th17cells in MM patients are significant higher thanthose in health controls，and the levels of Th17cells shows positive correlation withISS stage、β2-microglobulin and efficacy of MM patients。We conjecture that Th17cells may play an important role in the occurrence and development of multiplemyeloma，and it would contribute to judge the clinical course、efficacy and prognosisof MM patients。Maybe we can enhance the curative effect of MM patients throughtake Th17cells as a new therapeutic target of MM。...
