Effect Of L-methioninase On Gastric Cancer Cell Proliferation

Objective: TO study the effect of L-methioninase on proliferation of gastric cancercell line AGS.And investigate that is there the synergistic effect between Oxaliplatinand L-methionibase.Methods:1.After treated by L-methioninase or L-methioninase and Oxaliplatin, gastric cancercell were surveyed by MTT assay to determine cell proliferation.2. After treated by L-methioninase or L-methioninase and Oxaliplatin, We use flowcytometry to observe the distribution cell line when gastric cancer cell were treatedby L-methioninase or L-methioninase and Oxaliplatin.Results: Our study found that L-methioninase could inhibit the proliferation ofgastric cancer cell line AGS,and then induced apoptosis. L-methioninase lead cellgrowth inhibition and apoptosis in a time and dose dependent manner.The greasteffect appeared72h after L-methioninase(0.6Unit/ml)was administered.And the ratioof apoptosis of experimental group was significantly higher than the controlgroup(50.28%±6.84%vs12.95%±8.93%, P <0.05). Our study also found thesynergistic effect between Oxaliplatin and L-methionibase. WhenL-methioninase+Oxaliplatin were administered,the ratio of apoptosis significantlyhigher(63.73%±8.72%vs49.26%±8.57%, P <0.05).Conclusion:1.Our study found that L-methionibase could inhibit the proliferation of gastric cancercell line AGS.
