Background A variety of malignant tumors in China, the incidence of gastric cancer is the first. Postoperative metastasis and recurrence rate was high. Oxaliplatin is a third-generation platinum anti-cancer drugs, it has been widely used in the course of chemotherapy in gastric cancer. Studies have shown that tissue factor is not only widely expressed in human normal cells, but also high expression in a variety of tumor cells, and in tumor cell invasion and metastasis has played an important role.Objective To observe the role of tissue factor(TF)on Oxaliplatin ininhibits invasion and induced apoptosis in human gastric cancer cell.Methods Using human gastric cancer cell line SGC7901 wich has been successfully transfected with TF-pcDNA3.1 recombinant(Experimental group) and empty vector pcDNA3.1 (Control group) incubated with oxaliplatin.Transwell chamber were used to show the change of invasive ability of cells.Caspase-3 activity were evaluated by colorimetric method; Detection of oxaliplatin cytotoxicity by WST-1 method.Results1. Comparison of cell invasiveness of two groups:Comparison of cell invasiveness of two groups,the nuclei on the polyester film surface of Transwell chamber is more than the control, P<0.05.2. Comparison of Caspase-3 activity of two groups:Caspase-3 activity of experimental group were lower than the control group, P<0.05.3. Cytotoxicity results:The cell survival rate of experimental group was higher than the control group, P<0.05. Conclusion Increased expression of tissue factor can reduce the oxaliplatin on human gastric cancer cell invasiveness in vitro inhibitory effect and reduced oxaliplatin-induced apoptosis.
