Objective: The experiment establishes the model of COPD by the passive smokingand LPS dropped into the trachea. Observe the difference of pathology, the changes ofultrastructure and index of correlation in lung and heart of rats. Meanwhile analyses“relation of lung and heart” essentially by theory, literature and clinical.Method: The experiment divides32SPF healthy male Wistar rats into three groupsrandomly: the blank group (A), the COPD group (B), and the acute exacerbation of COPDgroup (C),8rats in each group. Then establish the model of COPD by the passive smokingwith LPS dropped into the trachea. Based on the model of COPD, drop the LPS into thetrachea again to establish the acute exacerbation model of the COPD. The blank group ratsraise in smoke-free environment. Observe general condition of rats, such as skin, hair,breath, response to stimulus, etc. Then collect the samples. Observe the organizationstructure of the lung and the heart by the light microscopy and electron microscopy. Detectthe content of the TGF-β1, CGRP in lung and heart, cTnI in serum with the ELISA, theexpression of the anti-M2receptor mRNA in blood with the RT-PCR. Statistics processexperimental data using SPSS18.0software.Result: research results make it clearly. The TGF-β1were expressed in the lung andheart tissue of the experimental rats,which in lung increased progressively in the COPDgroup and the acute exacerbation of COPD group(P<0.05), especially in the COPD acuteexacerbation increased obviously, which had more statistical significance compared withthe normal group (P<0.01). The TGF-β1in heart was not statistical significance comparegroup A with group B. The TGF-β1in group C had more statistical significance comparegroup A with group B(P<0.01);CGRP expressed in the lung and heart tissue of the experimental rats,the highest in group A, group B take second place(P<0.05)CGRP inlung and heart of group C expressed flat obvious(P<0.01); cTnI in the group of Cexpressed highes(tP<0.01), It was not statistical significance compare group A with groupB. the expression of the anti-M2receptor mRNA in group B was higher than group A(P<0.05).Group C was higher than group A and group B. The lung tissue appeared inorder inflammatory cells assault bronchial round, necrotizing inflammation in bronchus,congestion anf hyperplasia in pulmonary intersitital, multifocal necrotizing inflammation,lung congestion etc. Myocardial cells appeared in order eosinophilic change, adipose cellhyperplasia, interstitial osteoporosis disease in light microscopy. The structures of lung andheart changed differently in different times of COPD. Degree of injury was related tocondition of COPD.Conclusion:The experiment is established in Pulmonary disease spreading to heartand confirms relevance in lung and heart by Theory and clinical. announces theconnotation and essence of “Pulmonary heart related” in a certain extent., so as to supplysome ways or gist for developing the research and applied scope for this theory. Observethe development process of COPD by microcosmic and macroscopic.supply a new way toprevent treatment of COPD. It has positive meaning in preventing treatment of COPD.
