Objective: Human Fascin is a important member of the Fascins family,it is acytoskeletal protein,it mainly located in filaments and flake pseudopodia andcells spinous process.Fascin contains two actin binding sites,which regulatethe binding activity of Fascin binding with F-actin-binding and the formationof the filopodias,and so on.The functions of Fascin contain regulate migrationand adhesion of cell,and intercellular communication,et al.The up-regulatedexpression of it has been found in almost all of the cancer cells,overexpression of Fascin has a poor prognosis.P27is amember of the Kipfamily,it is a negative regulator of the cell cycles,it through inhibitecyclinE-CDK2compounds to arrest cell cycle in the G1phase and achieve theeffect of blocking cell proliferation,it can regulate cell cycle and induceapoptosis and many other functions.P27is related to the changes of celladhesion and cell migration.The down-regulated expression of P27can befound in a variety of malignant tumors,and may be associate with theinvasion,metastasis and prognosis in malignant tumors.Through the researchof Fascin and P27at home and abroad,revealed it associated with themovement,adhesion and proliferation of tumor cells,its molecular mechanismremains unclear,and the study of the relationship between them isminimal.Application of immunohistochemical staining methods,detect expression of Fascin and P27in colorectal cancer and normal tissue as well asadenomas.Analysis of their relationship with biological behavior of colorectalcancer and clinicopathological factors,and preliminary study of the correlationbetween them.Methods: We selected colorectal cancer tissues in60cases from January2011to April2012at department of General Surgery,Affiliated Hospital ofLuzhou Medical College.The cases were diagnosed as primary colorectalcancer and did not have any adjuvant therapy before surgery,andpostoperative specimens of all colorectal cancer diagnosed by pathologicalexamination.All specimens had intact cancer tissues and normal cutting edgetissuesand fetched60tissues at normal cutting edge of60cases respectivelyas normal tissue specimens.Another35adenoma specimens took frompatients who accepted surgical or colonoscopic resection of colorectaladenoma.All cases had complete clinical and pathological datas.Byimmunohistochemical SP method,detect expression of Fascin and P27proteins in60cases of cancer tissue specimens,Analysis of their relationshipwith the pathologic type,Dukes stages.And compared with the expression ofFascin and P27in60cases of normal cutting edge tissues with in35cases ofcolorectal adenoma specimens.Results: The positive expression of Fascin in colorectal cancer had32cases,the positive expression rate was53.3%,28cases were the negativeexpression.The positive expression of Fascin in adenoma had9cases,the positive rate was25.7%,26cases were the negative expression.All were thenegative expression of Fascin in normal tissues, the positive rate was0%.Compared with the positive expression of Fascin in cancer and adenomawas significant differences (P=0.009),Compared with the positiveexpression in cancer and normal tissues was significant differences (P=0.000),the positive expression of adenoma compared with normal tissues wassignificant differences (P=0.000).The positive expression of P27incolorectal cancer was26cases,the positive rate was43.3%,34cases wasnegative expression.P27expressed located at the nucleus and cytoplasm incancer.The expression of P27in adenoma was19cases,the positive rate was54.3%,16cases was negative expression.The expression of P27in normaltissues was47cases,the positive rate was78.3%,and the negative expressionwas13cases.The expression of P27was mainly expressed located at thenucleus in adenoma and normal tissues.Compared with the positiveexpression of P27in cancer and adenoma was no significant differences(P=0.302).Compared with the positive expression Of P27in cancer tissue andnormal tissue was significant differences (P=0.000),Compared with thepositive expression Of P27in the adenoma and normal tissues was significantdifferences (P=0.014).The expression of Fascin in colorectal cancer wasrelated to degree of differentiation,Dukes stages and depth of invasion andlymph node metastasis,has nothing to do with age,gender,tumor site,tumorsize.The expression of P27in colorectal cancer was related to Dukes stages and degree of differentiation,lymph node metastases,has nothing to do withgender,age,tumor size,tumor site,depth of invasion.The expression of Fascinand P27in colorectal cancer was negative correlation (r=-0.733, P=0.000).Their expression in adenoma was also negative correlation.Conclusion:1. In colorectal cancer,the expression of Fascin upregulated has nothingto do with gender,age,tumor location,and size,it is related to the degree oftumor differentiation,Dukes stage,depth of invasion and lymph nodemetastasis.2.The expression of P27downregulated in colorectal cancer is related toDukes stage,degree of tumor differentiation and lymph node metastasis,andhas nothing to do with gender, age, location, tumor size and depth of invasion.3. The expression of Fascin is negatively related to the expression ofP27in colorectal cancer and colorectal adenoma,and their expression isrelated to the degree of tumor differentiation,lymph node metastasis andDukes stage,Fascin and P27in the development and progression of colorectalcancer may play a certain synergy.4.Fascin and P27can be used as a potential indicators of colorectalcancer clinical diagnosis and prognosis.5.Regulate the expression of Fascin and P27,Suppresses the shift of P27expression localization,may provide a new strategy and the basis to thetreatment of colorectal cancer.
