Objective: To investigate the expression of ZEB1in colorectal cancertissues and four kinds of colon cancer cells with different invasiveness andmetastasis. At the same time Claudin1and E-cadherin were detected incolorectal cancer tissues. Methods:The expressions of ZEB1in differentinvasive potential colon carcinoma cell lines(HCT116, SW480, SW620,LOVO cell) were detected by PT-PCR and Western blot.51casesspecimens of colorectal carcinoma and para-tumor tissue were examed forZEB1, Claudin1and E-cadherin with the method of immunohistochemistry.The relationship among the expression of ZEB1, Claudin1and E-cadherinand clinicopathological significance were also analyzed. Results: ThemRNA and protein expressions of ZEB1were significantly higher in highinvasive potential cells (LOVO cells and SW620cells) than in low invasivepotential cells (HCT116and SW480cells), and the difference is significant(P<0.05). The positive rate of Claudin1and ZEB1in colorectal tumortissues (68.6%,23.5%) is higher than in normal colorectal mucosa(37.3%,0), with a significant difference between the two groups (P<0.01). Theexpression rate of Claudin1and ZEB1in III-IV stage (87.5%,41.7%) was higher than I-II stage (51.9%,7.4%;P<0.01,P<0.01), as in the cases withlymph node metastasis (90.9%,45.5%) than without metastasis (51.7%,6,9%;P<0.01,P<0.01). The expression rate of ZEB1in high grade (42.1%) wasalso higher than low grade (12.5%, P <0.05). The expression rate ofE-cadherin in I-II stage (74.1%) was also higher than III-IV stage(37.5%,P<0.05),as in the cases with no lymph node metastasis (72.4%)than lymphnode metastasis (36.4%, P<0.05).Positive correlations were observedbetween Claudin1and ZEB1in CRC tissues(r=0.358, P<0.05). Theexpression of E-cadherin was negatively correlated with the expressions ofClaudin1and ZEB1(r=-0.302, r=-0.357;P<0.05,P<0.05).Conclusion:ZEB1was found to be closely related to epithelial-mesenchymal transitionin colon cancer cells. Expression of Claudin1, ZEB1and E-cadherin may besignificantly involved in the progression, invasion and metastasis of CRC.Claudin1may up-regulates ZEB1to inhibit E-cadherin expression.Claudin1, ZEB1and E-cadherin may promote CRC progression.
