Objective: To observe the inhibitory action of the spleen cytotoxic Tlymphocytes(CTL) obtained from immuned mice by dendritic cell vaccinecarrying HPV-16E6/E7gene on cervical cancer in nude mice and Studiesthe related mechanisms.Methods: Immature C57BL/6DC (imDC) was culture and wasinfected with recombinant adenovirus vector carrying humanpapillomavirus-16E6/E7gene.Expressions of E7protein were detected byWestern blot. CaSki cell transplantable tumor model of human cervicalcancer was constructed in nude mice,and was treated by cytotoxic Tlymphocytes (CTL),acquired from BALB/c mice.Changes in tumor volumewere observed.Nude mice were sacrificed and Paraffin-embedded at the28th d since tumors appeared.Tumor cell morphology was observed in eachgroup by HE staining. Expressions of Bcl-2and Bax protein of tumortissues were detected by immunohistochemical techniques. The proliferation of spleen T lymphocyte of BALB/c mice immunized by DCvaccine were detected by CCK8(cell counting kit8).Expression of CD4﹢of spleen tissue of nude mice.Results: DC was successfully cultured and DC vaccine wasprepared.At the28th d,tumor volume of nude mice was smaller inpAd-E6E7-DC-CTL-CaSki group than in pAd-mock-DC-CTL-CaSkigroup, DC-CTL-CaSki group and CaSki group (P<0.05].Bax proteinexpressions of tumor tissues detected by immunohistochemical techniquesdemonstrated that:expressions in pAd-E6E7-DC-CTL-CaSki group werethe highest while in CaSki group were the lowest.Bcl-2protein expressionsof tumor tissue detected by immunohistochemical techniques demonstratedthat:expressions in pAd-E6E7-DC-CTL-CaSki group were the lowest whilethose in CaSki group were the highest. the OD values of T lymphocytes ofpAd-E6/E7-DC group were significantly higher than the pAd-mock-DCgroup、DC group and PBS group (P <0.01).The expression of CD4﹢washigher in pAd-E6/E7-DC-T-CaSki group than in pAd-mock-DC-T-CaSkigroup, DC-T-CaSki group and CaSki group.Conclusion: The DC vaccine which were modified by HPV-16E6/E7gene, can promote the proliferation of T lymphocytes in vivo, andinhibit the grow of cervical cancer transplantation tumor in nude mice byinduce CTL..
