ObjectiveTo investigate the pathogenesis of IL-17、MMP-3on rheumatoid arthritis(RA),and to observe the effect of Recovery and Benefiting kidney(R&B)soup on the expression of pathology change and its cytokines in synovialtissue of type Ⅱ collagen induced arthritis (CIA) rats; and furtherinvestigate the possible mechanism of R&B soup treatment effect on RA.MethodRA rats were induced by cattle collagen Ⅱ combined with cold-wetnessfactors, which were divided into normal group, model group, methotrexategroup, each R&B soup groups(high dosage, middle dosage, low dosage)and combination group(high dosage of R&B soup with methotrexategroup).On the4th,8th,12th week, we calculhated the rat feet swellingrate,and detected serum TNF-α and IL-17level by enzyme linkedimmunosorbent assay (ELISA), MMP-3level byimmunohistochemistry(IHC), rat feet joint synovial tissue pathologicalchanges by light microscopic after the12th week. ResultIn the model group, they moved very slowly with red and swollensignificantly feet, while their Mankin’s scoring, serum TNF-α and IL-17level, and MMP-3level were increased significantly (P<0.05), the longertreatment the better (P<0.01). Compared with model group, after4w’smedication, the serum TNF-α level was decreased in high, middle dosageof R&B soup groups, MTX group, and combination group(P <0.05); andthe level of serum IL-17was decreased in MTX group and combinationgroup (P <0.05); but the knee cartilage and synovial layer MMP3in all thegroups did not change (P>0.05). Medication8w later, the serum TNF-αlevels were decreased significantly (P<0.01) in high, middle dosage ofR&B soup groups, MTX group, and combination group (P<0.01); in MTXgroup, in high dosage of R&B soup group and combination group with aserum level of IL-17have decreased significantly (P<0.01); the articularcartilage and synovial layer MMP3level were also decreased in MTXgroup, Chinese high-dose group(P<0.05). After medicationed12w, theserum TNF-α level was decreased in all the groups (P <0.01); while theserum IL-17level and knee cartilage and synovial layer of the MMP3were decreased significantly in MTX group, Chinese high dose group, andombined with water average (P<0.01). Which, to the role of R&B souphigh-dose group and MTX group, no statistically significant difference (P>0.05), but the combination group was superior to the other two groups, there was a statistically significant difference(P<0.05).Chinese medicinehigh-dose group and the combination group can significantly improve RAsynovial tissue of rats pathology form to promote the absorption ofsynovial inflammation to reduce synovial hyperplasia and improvecartilage and bone destruction, close to the combination group and thenormal group.Conclusion(1)It is the positively correlated relationship among TNF-α, IL-17andMMP3. It explained that IL-17, MMP3were the main medium in RAjoint injury and played an important role in the pathogenesis of RA.(2)R&B soup can obviously improve the symptom of the CIArats, thereis no difference between the high dosage of R&B soup group and MTXgroup, which the mechanism is closely related to inhibit the releasing ofinflammatory mediators such as TNF-α, IL-17and MMP3, the longerduration of treatment the better, and the combination group is best. Itindicated that Chinese combined with western medicine treatment ofrheumatoid arthritis is outstanding.
