Objective: TO investigate the effect CQMUHS-03, a pioglitazonederivation, on proliferation and differentiation of3T3-L1. TO study themechanism of PPARγ agonist improving insulin resistance, provide theorybasement for clinical therapy diabetes and research anti-diabetes drugs.Method:(1)To detect different concentrations of CQMUHS-03atdifferent time on3T3-L1proliferation by MTT.(2) To measure differentconcentrations of CQMUHS-03at different time on3T3-L1differentiationby oil red dyeing.(3) Western Blot was introduced to detect the expressionof TNF-α in3T3-L1treated with CQMUHS-03during the differentiation.The expression of adiponectin and C/EBPα mRNA in differentiation of3T3-L1treated with CQMUHS-03were evaluated by RT-PCR.Results:(1) MTT showed that CQMUHS-03and standard pioglitazoneinhibited3T3-L1proliferation in a dose and time dependent manner, andCQMUHS-03had a weaker function than standard pioglitazone.(2)CQMUHS-03and standard pioglitazone could promote3T3-L1differentiation in oil red dyeing results, but the effect of standardpioglitazone was obvious.(3)The expression of TNF-α protein was modulated during the time in3T3-L1differentiation. CQMUHS-03andstandard pioglitazone decreased more at2d and8d than4d and6d. WhileCQMUHS-03expressed less than standard pioglitazone. The RT-PCRshowed at8th day of differentiated3T3-L1, the expression of adiponectinand C/EBPα mRNA increased in CQMUHS-03and standard pioglitazonecompared with control group. The level of adiponectin mRNA was higher inCQMUHS-03than standard pioglitazone, whereas standard pioglitazone hadhigher expression of C/EBPα.Conclusion: CQMUHS-03could improve proliferation anddifferentiation of3T3-L1. The mechanism may include in the mRNAexpression of PPARγ and C/EBPα, which promote differentiation of3T3-L1.This study approves evidence and theory guidance for clinical treatment ofdiabetes and other metabolic diseases using thiazolidinediones.
