Objective: The model of hyperlipidemia rats were established by high fat diet toinvestigate the effect of hyperlipidemia on pancreatic cell in rats.Methods:30SD rats were randomly divided into three groups. There was10rats ineach group. Control group were fed for normal diet, while group1and group2werefed for high fat diet I and II during45days to establish thehyperlipidemia model. In45days group2were injected to intraperitoneal for75%egg yolkemulsion, but group1group and control group injectedvolume0.9%NS. Blood cholesterol (TC), triglyceride (TG), high densitylipoprotein (HDL-C), low density lipoprotein (LDL-C), serum amylase (AMY), andserum tumor necrosis factor alpha (TNF-α) were observed, and free fatty acids(FFA)and the expression of Caspase in pancreas were measured. Pancreatic tissue pathologyand pancreatic ultrastructure (TEM) were inspected in rats after10hours.Results: Compared with the control group, TC,TG,,HDL-C,LDL-C, serum AMYand TNF-α, the pancreatic FFA, the expression level of Caspase in the pancreas,pancreatic pathological evaluation in the group1and group2was significantlyincreased (P<0.01). There were appeared mild edema of pancreatic interlobular,focal bleeding of a small amount, a small amount of chronic inflammatory cellinfiltration and the pancreatic acinar cellsshowed dilation of rough endoplasmicreticulum in group1and2. TC, TG, HDL-C, serum TNF-α and AMY and LDL-C,the pancreatic FFA, pancreatic pathological score and pancreatic tissue Caspaseexpression level in group2was significantly increased than one in Groups1(P<0.01,P<0.05),but in group2part degeneration and necrosis of thepancreas tissue, pancreatic interlobular edema, focal hemorrhage, chronic inflammatory cell infiltration, pancreatic acinar cell rough endoplasmic reticulum expansion andsome mitochondria appearedmild swelling were observed respectively bypathology and electron microscope. The relation between TC,TG and the pancreatic FFA levels,TNF-α were respectively positively correlated (r=0.8404and0.6914;r=0.9337and0.8249).Conclusion: High fat diet and intraperitoneal injection of egg yolk emulsion cansuccessfully induce the animal model of hyperlipidemia; hyperlipidemia cancause injury of pancreatic cells, it may be importance reason that acute pancreatitismay tend to be induced in hyperlipidemia.
