Objective:To study on the expression of interleukin17(IL-17) and interleukin17receptor (IL-17R) in different grades of glioma tissue and to evaluate the relationshipbetween its expression and the pathological grades of glioma. Methods:(1) Groupdivision:In this research,31patients with glioma were studied(WHO histological gradeⅠin2,gradeⅡin10,gradeⅢ in9,gradeⅣ in10)as the experimental group. They weredivided into high-grade gliomas(including WHO Ⅲ&Ⅳ)and in low-grade glioma(including WHO Ⅰ&Ⅱ).10patients with craniocerebral trauma underwentdecompressive craniotomy as control group. Meanwhile, peripheral blood samples werecollected in the31patients with glioma and10cases with craniocerebral trauma.10casesof healthy volunteers were divided into the control group, peripheral blood samples werecollected.(2) The immunohistochemical detection used to stain IL-17and IL-17R inhigh-grade gliomas, low-grade glioma and the control group specimens respectively, therelationship between IL-17and IL-17R and pathological grades of glioma wasanalyzed.(3)The real-time fluorescence quantitative PCR method was used to detect theexpression of mRNA level of IL-17and IL-17R in different grades of glioma specimens,peripheral blood of patients with different grade gliomas and the control group, therelationship between IL-17and IL-17R and pathological grades of glioma was alsoanayzed.(4)The protein immunoblotting method used to detect the expression of proteinlevel of IL-17and IL-17R in different grades of glioma, peripheral blood of patients withdifferent grade gliomas and the control group, the relationship between IL-17and IL-17R and pathological grades of glioma was anayzed. Results:(1) The result ofimmunohistochemical detection showed that the portrition of positive expression of IL-17in control group, low-grade gliomas and high-grade gliomas were30%,67%,100%respectively, the mean rank order were16.84,31.31,45.43accordingly, there was asignificant statistical difference in the expression of the general (HC=24.63, P <0.05), andthe expression of the positive rates of IL-17R in control group, low-grade gliomas andhigh-grade gliomas were50%,83%,100%, the mean rank order is respectively18.42,35.73,49.65, there was a significant statistical difference in the expression of the general(HC=28.42, P <0.05). IL-17R in high grade glioma tissues was higher than that in lowgrade glioma tissues(p<0.01), and both were higher than the control group(p<0.05).(2)The result of Real-time PCR method showed that the expression of IL-17mRNA in controlgroup, low-grade gliomas and high-grade gliomas were31.62±1.95,85.79±4.58,134.77±6.32, and IL-17R mRNA in these were76.20±3.97,137.66±64,193.82±6.20. Bythe analysis of variance (F=221.04, p<0.05).There were significant difference betweenevery two groups. The expression of IL-17mRNA in control group, low-grade gliomas andhigh-grade gliomas patients peripheral blood were42.09±1.65,48.21±1.79,103.71±4.37,184.67±6.51, and IL-17R mRNA in these were83.18±4.57,89.47±3.82,158.45±5.34,237.32±7.29. By the analysis of variance (F=274.64, p<0.05). There was no significantdifference between the healthy control group and decompression group (t=-0.92,p>0.05),but other groups had statistical significance.(3) The result of Western blot methodshowed that the protein level of IL-17in control group, low-grade gliomas and high-gradegliomas tissues were0.23±0.01,0.42±0.01,0.67±0.02, and IL-17R in these were0.34±0.01,0.68±0.02,0.89±0.03.By the analysis of variance (F=89.23, p<0.05), there weresignificant difference between every two groups.The protein level of IL-17in controlgroup,low-grade gliomas and high-grade gliomas tissues were0.33±0.01,0.35±0.01,0.54±0.02,0.85±0.02,and IL-17R in these were0.42±0.01,0.43±0.01,0.78±0.02,0.94±0.03. By the analysis of variance (F=107.89, p<0.05). There was no significantdifference between the healthy control group and decompression group(t=-0.83, p>0.05),but other groups had statistical significance.(4) A significant positive correlation between the expression of IL-17in peripheral blood of patients with the same tumor andcorresponding glioma tissues (r=0.520, p=0.003) was revealed, and the IL-17R expressionin venous blood and tumors have the same significant positive correlation.(r=0.636,p<0.001). Conclusions: The expression intensity of IL-17and IL-17R and pathologicalgrading of glioma are closely related. There were differences in the expression of IL-17and IL-17R in the peripheral blood and patients with different grade glioma, and they wentup with the increase of the glioma grading. Significant positive correlation was foundbetween the expression intensity of IL-17and IL-17R in the peripheral blood and gliomatissue from same patient. It can be concluded that IL-17and IL-17R can be used as areference factor to judge the malignant degree of glioma.
