Effects Of Duodenal-jejunal Bypass (DJB) Surgery On GLUT-4in Type2Diabetic SD Rats

Objective: To discuss the relationship between DJB surgery and GLUT-4in skeletalmuscle and myocardial of T2DM SD rats.Methods: Forty eight-week SD rats were randomly divided into four groups: type2diabetes-operation group(To group) and type2diabetes-operation group(No group),normal-operation group(No group) and normal-control group(Nc group). The weight,fasting blood glucose, insulin and the expression of GLUT-4protein were measured andrecorded respectively before operation and1week,2weeks,4weeks as well as8weeksafter operation in all rats. Pathological changes in islet tissue were observed undermicroscope after HE staining.Results:①The weight loss in To group rats was meaningless(P>0.05).②At8weeks after surgery the fasting blood glucose of To group rats decreased to4.05±2.39mmol/L from22.71±5.26mmol/L at pre-operation(P<0.05).③the fasting bloodinsulin of To group rats decreased to6.35±3.38mU/L from11.61±0.51mU/L(P<0.05),and close to preoperative value of insulin.④the GLP-1in serum of To group rats from2.18±0.56pmol/L upto11.16±5.92pmol/L(P﹤0.001), and close to preoperative value ofGLP-1.⑤Tc group islet has obvious pathological changes, but To group are close to the No,Nc group, and has a tendency to be gradually returned to normal.⑥The expression ofGLUT4protein in skeletal muscle and myocardium significantly increased.Conclusion: Though DJB surgery can’t reduce the weight of T2DM SD rats, thissurgery can lower blood glucose and improve insulin in blood and enhance theexpression of GLUT4protein in skeletal muscle and myocardium of T2DM SD rats.
