Objective: This research adopts the model of multiple organ dysfunction syndrome(MODS) with landrace,to explore the therapeutic effect and mechanism with traditionalChinese medicine of Xuebijing of prevention and treatment on Highland multiple organdysfunction syndrome (H-MODS),to provide experiment evidence for clinical application.Method: Choose24healthy male landrace with the weight of10~15kg and2months,after adaptive feed1week, rapidly sent to Sanke Grasslands (elevation3200m) fromLanzhou(elevation1520m) to do the experiment, After all the landrace wereanesthetic,randomly divided into blank control group (group A),model group (group B), thelow dose group of Xuebijing (group C), the high dose group of Xuebijing (group D), eachgroup with6landraces. the Group B,C,D landrace via ear marginal vein driplipopolysaccharide(LPS),0.35mg/kg,making the model of MODS,LPS was finished, C groupwas given:the Xuebijing with1.5mL/kg,2/day;D group was given:Xuebijing with3ml/kg,2/day;A and B group was given:such as physiological saline volume,2/day; For3days.In different time to observ the changes of vital signs,viscera functionindex(ALT,AST,TBIL,CR),flammatory factors(TNF-α,IL-1) and the viscera pathologicalchanges of every groups of landraces;Contrast the statistical differences of everygroups andthe change rules of each time in the group, draw a conclusion.The experimental dataapplication SPSS17.0statistical analysis software package for analysis.Results:1.B, C, D groups of body temperature, respiration, heart rate increasedsignificantly,compared with group A, the difference was statistically significant (P<0.05orP<0.01);Group B breathing, heart rate higher than C and group D, the difference wasstatistically significant (P<0.05, P<0.05);24h group B33.3%mortality rate, C, D groupsmortality rate was16.6%.2. B, C, D groups of white blood cell (WBC) are “Scoop”typechange. Group C, D the WBC rise range was lower than group B(P<0.05or P<0.01). GroupD the WBC rise range was lower than group C in48h(P<0.05).3. The AST, ALT, TBIL,CR ofB, C, D groups rapidly rising in6h,48h a peak, The most obvious rising was group B,compared with group C and group D, the difference was statistically significant (P<0.05orP<0.01),after gradually declined. After24h,group D the AST,CR rise range was lower than group C(P<0.05, P<0.05).4. Compared with group A, B, C, D groups of TNF-α,IL-1expression higher in6h(P<0.01),reached a peak in12h(P<0.01).The most obvious rising wasgroup B, compared with group C and group D, the difference was statistically significant(P<0.05or P<0.01), after gradually declined.Group D the TNF-α in24h and the IL-1in48hrise range was lower than group C, the difference was statistically significant (P<0.05,P<0.05);B, C, D groups NF-κBmRNA expression was obviously higher than that of group Ain24h(P<0.01), The most obvious rising was group B.in72h, group B the NF-κBmRNAexpression was still higher than group A(P<0.05),but group C and D the NF-κBmRNAexpression was close to the level of group A.Conclusions: The intervention treatment with Xuebijing in model of LPS inducedHighland landrace MODS, not only reduce the mortality rate of H-MODS model landrace,hepatic and renal function injury caused by endotoxin, but also block the release ofinflammatory mediators, to reduce the stimulation of inflammatory factor in the body,improving the survival rates of H-MODS, It shows that Xuebijing have good prevention andtreatment to H-MODS.
