Objective:Observing the effects of expression of (phosphatidylinositol-3-kinase, PI-3K) and (glucose transporter4, GLUT4) in type2diabetic rats skeletalmuscle tissue,using Chinese Patent Medicine Huazhuo Granules. Preparation of ratmodel of type2diabetes, use Chinese medicine decoction made of Huazhuogranules to intervene T2DM rats,and to investigate whether Huazhuo granules canaffect the activation of PI-3K and GLUT4signaling pathways, thereby improvinginsulin resistance, to investigate the molecular mechanism of prevention of type2diabetes.Verification the method of detoxification and removing turbidity can improvethe scientificity and rationality of insulin resistance. Also provide experimental basisfor the development of Chinese medicine insulin sensitizers, and thus lay thefoundation for the prevention and treatment of traditional Chinese medicine ormethod of integrative medicine.Methods:Select60SPF level of healthy, male wistar rats (4months,200±20g),adaptive feeding one week, to fed the basal diet, and randomly selected12rats as thecontrol, and the remaining48as a model group, after high-fat diet for four weeks,using this high fat and sugar combined with low-dose streptozotocin (streptozotocin,STZ,30mg.kg-1continuous administration of3days) intraperitoneal injection methoddiet method, to build2type diabetic rat model, at72h after, not provide diet but water,rats was measured fasting blood glucose and OGTT2hours blood glucose through thetail vein of rats.If FBG72h after modeling rats≥7.0mmol/L,OGTT(2hPG)≥11.1mmol/L, is the successful model. The successful model rats wererandomly divided into Huazhuo granules group(2.025g/kg), rosiglitazone (0.36mg/kg),model group (1ml/100g), continued high-fat diet feeding, and be with the controlgroup(1ml/100g) together consecutive treatment, the model group and the controlgroup were to be saline, observe the general state of the rats, body weight and foodintake,10w after the intervention, intraperitoneal injection10%chloral hydrate toanesthetized rats, test FBG,OGTT(2hPG),FINS level, and apply the method of reversetranscription polymerase chain reaction(RT-PCR) to detect the skeletal muscle tissueof rats PI-3K and GLUT4mRNA gene expression levels.All data using SPSS19.0software for statistical analysis.Results:1. Weight of high fat and sugar diet groups was significantly increased compared with normal diet group, the difference was statistically significant (P<0.05), However,there is no significant difference between fasting glucose of high-fat group comparedto the normal diet group(P>0.05), if you give a small dose of STZ (30mg.kg-1) to thehigh-fat diet groups,after intraperitoneal injection,the blood glucose levels of high-fatdiet group was significantly elevated, compared with the basal diet group, thedifference was statistically significant(P<0.05); in this experiment,we use48rats andmake36rats models finally,suggesting that we successful replication T2DM animalmodels;2. After the drugs intervention10weeks, weight of rosiglitazone group and themodel rats were significantly increased compared with the control group(P<0.05), theweight of Huazhuo Granules group is less than model group, the difference wasstatistically significant(P<0.01),It suggested that Huazhuo Granules weight of obeseT2DM rats have lower effect of growth;3. Comparison with the control group of rats and model rats,FBG,2Hpg,FINSlevels were increased, and the difference was statistically significant(P<0.05),Huazhuo group and rosiglitazone group both can decrease FPG,2hPG and FINS level,comparison with the model group, the difference was statistically significant(P<0.05);there was no significant difference between the Huazhuo group and rosiglitazonegroup(P>0.05);4. The results of RT-PCR shows: the expression level of PI-3K and GLUT4inskeleton tissue of Huazhuo particles group and rosiglitazone group is significantincreased(P<0.05).However, in the control model, the level of PI-3K and GlUT4islow(P<0.05).Conclusion:1. Huazhuo granules through improving insulin sensitivity, improve insulinresistance in T2DM;2. Huazhuo granules can improve the level of PI-3K and GLUT4in skeletonmuscle tissue. Huazhuo particles can activate PI-3K and GLUT4in insulin signalpathway and increase the sensitivity of insulin.
