Objective:To observe the effect of TuoGuanSan to clear the Staphylococcus aureus andpromote wound healing,then to analyze the possible mechanism of action.Methods:1、Preparation of the standard bacteria:The Staphylococcus aureus were subcultured andpreparation the standard bacteria press1×109cfu/ml.2、Establishment of animal models, animal grouping and processing:To establish themodel of108healthy male Wistar rats with bacterial infection of skin defects and hormonalintervention.These rates were randomized to A group (experimental group,n=36), Bgroup(positive control group,n=36)and C group(model group,n=36).A group:To bound thewound with the gauze(3×4cm) which was soaked by1ml (0.005g/ml) TuoGuanSansuspension;B group to bound the wound with the gauze(3×4cm) which was soaked by1ml(0.4%)liquor iodi mitis.C group to bound the wound with the gauze(3×4cm) which wassoaked by1ml (0.4%)liquor iodi mitis.After dressing,to fix the gauze with the homemadesteel mesh.One time daily was needed to dress with the same way.3、Observations:To observe the general states of rats and wound healing time.The woundhealing rate and PH of wound secretions were observed after treatment for1,3,7,10,14,21days.Each group killed six rats randomly for quantification of subeschar bacteria,and takeanother part of tissues which come from the wound to observe the histomorphology by HEdying.The expression of epidermal growth factor (EGF) and vascular endothelial growthfactor (VEGF) were observed by immunohistochemistry after treatment for1,3,7,10,14,21days.Result:1、The activities、the dietary intake、the growth of acestoma、the inflammation andexudate around the scald wound of group A were more better than group B and C.2、 Compared the wound healing time of A group(18.92±1.08d)with Bgroup(20.78±1.39d)and C group(22.67±2.53d) was significantly less.There has a statisticallysignificant difference between A group and group B、C(P<0.05and P<0.01).Compared groupB and C,P<0.01. 3、The wound healing rate in each group: In the first3days and10days, group A、B havea statistically difference with group C(P<0.01);The wound recovery rate of group A washigher than group C(P<0.01)and B(P<0.05)in the first7days and10days;The wound healingrate of group A was also higher than group C(P<0.01).4.The PH of wound secretions:In day3,the wound secretions PH of A、B groups werelower than group C(P<0.01and P<0.05),There has a statistically significant differencebetween A group and group B(P<0.01);In day7,the wound secretions PH in group A wassignificantly lower than that in group B and C(P<0.01andP<0.05).5.The results of bacteriological test: At1,14days, the number of fungus of group A wasless than group C, there are significant differences between the two groups (P <0.05); At3、7、10days, A group and C group had a statistically significant difference (P <0.01), and therewere significant differences in group B and group C(P <0.05); At3days in group A andgroup B had a significant difference (P <0.01);There were significant differences betweengroup A and B at7、10days(P <0.05).6.Result of histopathological:After the first day treatment,the inflammatory responsewere significant in each group; Compared with group B and C, the wound exudate in group Awas more increased and the inflammatory cells、 fibroblasts and macrophage wereproliferation significantly.At7-10days, A group and B group were more granulation tissuefilling the wound.Compared with group B,the number and capillariesmore、the volume offibroblasts in A group were more higher.At14days,the number of fibroblasts weresignificantly reduced,New collagen fibers thick, began to appear a small amount of hairfollicle epithelium and other newborn skin appendages,Most of the wounds were covered bynewborn skin.After21days treatment,the wounds of group A and B were filled withgranulation tissue and a new skin,there were a large number of collagen fibers were arrangedin clusters or fascicles.7.The expression of EGF:The mean optical density in different time periods of EGF: At3、14days, there were significant differences between group A and group C (P <0.05);At14days, there were significant differences between group A and group B(P <0.05); At10days,group A and group C had a significant statistically difference (P <0.01); The average gray ofEGF expression in different time periods:At10、14days,there were significant differences between group A and group B、C(P <0.05).8.The expression of VEGF:The mean optical density in different time periods of VEGF:At3、7、10days, there were significant differences between group A and group C (P<0.01);At7、10days, roup A and group C had a significant statistically difference (P <0.05);The average gray of VEGF expression in different time periods:At10、14days,there weresignificant differences between group A and group B、C(P <0.05).Conclusion:1.TuoGuanSan can clear the Staphylococcus aureus in chronic wound.2.TuoGuanSan can reduce the pH of chronic wounds,3.TuoGuanSan adjust the expression of EGF、VEGF during wound healing4.TuoGuanSan maybe through acidate the wound, clear the wound bacteria andupexpression of EGF、 VEGF during the wound healing to promote the wound healing...
