Objective: It is to discuss the inhibited effect and mechanism of Diosbulbin B on theproliferation of the digestive system tumor cells in vitro. Methods: Human well differentiatedgastric cancer cell line MKN-28, moderately differentiated gastric cancer SGC-7901, poorlydifferentiated gastric adenocarcinoma BGC-823, Colon cancer LS-174T, Liver SMMC-7721,HEPG-2Hela cervical cancer activity were treated with various concentrations of DiosbulbinB for24h, then cell proliferation was detected using MTT assay.Determine the effect oncancer cell cycle distribution of Diosbulbin B by flow cytometry, then presume the actionmechanism of Diosbulbin B on tumor cells according to the cell cycle analysis results.Results: Diosbulbin B has different degrees of inhibition effect on human gastric cancer cellline MKN-28, moderately differentiated gastric cancer SGC-7901, poorly differentiatedgastric adenocarcinoma BGC-823, Colon cancer LS-174T, Liver SMMC-7721and HEPG-2Hela cervical cancer (P﹤0.05), When the Diosbulbin B at the concentration of40μmol/L,the inhibition rate of MKN-28and other5experimented kinds of cancer cells will reach up to30%after24hours. Conclusion: Diosbulbin B has a significant inhibition ability onproliferation and metastasis of MKN-28and other ten cells in vitro. Diosbulbin B, at theconcentration of40μ mol/L, has similar inhibition on cancer cells with cisplatin at theconcentration of20μ mol/L. thus, the effect on cell cycle distribution and inducing apoptosismay be attributed to the anti-tumor activity of Diosbulbin B.
