Objective: Analyzing the expression of TCR ζ chain and the related protein ZAP70inperipheral blood mononuclear cells (PBMCs) from dermatomyositis(DM) patients,in order toexplore the function of TCR/CD3complex in the pathogenesis of dermatomyositis. Methods:To assemble the peripheral blood and essential information of DM patients in our hospitalfrom Oct2011to Aug2012.Set. Real-time RT-PCR with SYBY Green Ⅰtechnique was usedfor detecting the mRNA level of TCR ζ chain and ZAP70gene in PBMCs of patients withDM and healthy adults served as normal control using relative quantification. β-actin wasused as an endogenous reference gene. To Analyze all data with SPSS (statistical program forSocial Science)17.0software. The compare of mean value use T-test. P<0.05meansstatistical difference. Results: The expression of TCR ζ chain and ZAP70mRNA in PBMCsfrom DM patients is significantly lower than those from normal control, P<0.05; In DMpatients,there is no difference between males and females, with arthritis or not, withinterstitial lung disease(ILD) or not,with weakness of muscle or not;The ζ chain mRNA levelof the DM patients with raynaud phenomenon is notably lower than those without raynaudphenomenon. There is no difference of TCR ζ chain and ZAP70mRNA expression levelbetween higher indexes (such as AST、Ck and LDH),ESR,CRP,Jo-1(+)and their own controls.Conclusion: In DM,the expression of TCR ζ chain and the related protein ZAP70isabnormal,lower than normal control,has some relation with raynaud phenomenon. And itmaybe one of the pathogenesis of T cell abnormal function in DM.
