Object: To study the effects of4-Hydroxyisoleucine (4-HIL) on the expression oftumor necrosis factor-α (TNF-α) in different concentrations and to explore thepossibility of using4-HIL to treat insulin resistance.Methods: Human adipocytes were treated with4-HIL of different concentration.Glucose uptake was assayed using2-[3H] DG. Protein levels were examined byELISA. Gene expression of TNF–α was measured by RT-PCR.Results:4-HIL increased glucose uptake in adipocyte with insulin resistance.Treatment of adipocyte with4-HIL of different doses (5,10,20μmol/L, respectively)significantly down-regulated TNF-α expression and mRNA of it when compared withthose treated with insulin alone or palmitic acid.Conclusion:4-HIL can increase glucose uptake and down-regulate TNF-α expressionin human adipocyte and thus inhibit insulin resistance. |
