Objective: To observe complex dragon’s blood mixtura on inhibiting renal fibrosis ofDiabetic Nephropathy Mice induced by STZ and to investigate the mechanism ofrenal fibrosis of Diabetic Nephropathy Mice.Methods: Mice were divided into5groups: normal control group, diabeticnephropathy control group. Diabetic Nephropathy in mice was induced by STZperitoneal injection. Two weeks later Mice were administed. The mice were killedafter treatment for four weeks. We detect the body weigh, kidney weight indexnumber, blood glucose and24hours Urine protein. The express levels of α-SMA、FN、 Nephrin and P-cardherin were detected by Western blot andimmunohistochemistry。Ultrastructural organization of kidney was detected bytransmission electron microscope。Results: As compared normal control group, the body weigh, kidney weight indexnumber and the express levels of Nephrin、P-cardherin were reduced significantly indiabetic nephropathy control group, and blood glucose,24hours Urine protein and theexpress levels of α-SMA and FN were increased significantly in diabeticnephropathy control group.Conclusion: Complex dragon’s blood mixtura can reduce the levels of DN mice urineprotein, reduce kidney weight/body weight index, inhibit matrix accumulation in renalcortex, and reduce glomerular basement membrane thickening. The mechanisms ofthese effects may be related with its down regulation the express levels of a-SMA andFN, and up regulation the express levels of nephrin and p-cardherin in DN mice renalcortex, that is to say, they may be related with its inhibition of EMT in DN mice renalcortex.
