Objective: To observe the effects of total flavonoids of salamander on differentconcentrations on the model of osteoporosis in ovariectomized rats (Ovariectomized Rats,OPGmRNA, OVX) of RANKLmRNA and RANKmRNA expression, and the serum Calcium(Ca2+), Phosphorus (P), Estradiol (E2) concentrations, and to explore the mechanism, for theclinical use of Chinese medicine salamander prevention and treatment of osteoporosis inpostmenopausal osteoporosis and provide experimental basis.Method: The osteoporosis in ovariectomized rat model as molding method, select SPFlevel of56healthy female SD rats in age of4monthes, according to the weight of the animalsubjects were randomly divided into7groups: sham operation group, model control group,positive control group, total flavonoids of salamander high, medium and low dose group,blank control group, with8rats in each group. The model group, positive control group, totalflavonoids of salamander high, medium and low dose group in the animal experimental centerof Gansu College of TCM SPF underwent bilateral ovariectomy, resulting to OVX. Normaldiet for3monthes, calculate the corresponding drug dosage after weighing, calculated inaccordance with the rat "human and animal body surface area equivalent dose" dose: the totalflavonoids of salamander equivalent dose for medium dose (210mg/kg),1/2and2timesrespectively for low dose (105mg/kg) and high dose (420mg/kg); equivalent dose ofAlendronate sodium in positive control group was1.05mg/kg, by gavage with double distilledwater to dissolve,3ml each time,1times/day, sham operation group, model control groupand blank control group and normal saline by intragastric gavage, the rats were sacrificedafter one month. Then to run the RT-PCR, relative quantitative real time detection of bonetissue OPGmRNA, RANKLmRNA, RANKmRAN expression, calculating ΔCt values and2-ΔΔCtvalues, the expression intensity of OPGmRNA analysis between groups,RANKLmRNA, RANKmRAN SPSS17.0, serum Ca2+, P, E2concentration in rats of eachgroup were detected prognosis dry, experimental data with the mean SD () said, amonggroups were compared with single factor analysis of variance, P<0.05has the remarkabledifference, the difference has statistical.Result: 1The total flavonoids of salamander intervented to OVX dose treatment could improvethe expression intensity of OPGmRNA in the bone tissue, RANKmRNA, compared to themodel control group, blank control group, high dosage group, low dosage group and positivecontrol group were significantly different (P<0.05), the difference has statistical significance.2The total flavonoids of salamander dose can reduce the intensity of RANKLmRNAexpression in bone tissue, compared with model group, blank control group, high dosagegroup, low dosage group and positive control group were significantly different (P<0.05), thedifference was statistically significant.3Effectively improve the serum Ca2+, P, E2dose can total flavonoids of salamander,compared with the positive control group, high dose group, low dose group concentration,medium dose group were significantly increased (P<0.05); compared with the model group,middle dose group, blank control group, sham operation group concentration have significantdifferences (P<0.05); the difference was statistically significant.Conclusion:1The dose intervention in OVX of total flavonoids of salamander can significantlyimprove the strength of OPGmRNA, RANKmRNA expression in bone tissue, the expressionintensity of RANKLmRNA can be inhibited bone tissues, bond between the inhibition ofRANKL and RANK OPG competitive, and inhibit the generation of mature osteoclasts,played anti osteoporosis the role of.2The dose drug intervention of total flavonoids of salamander could increase the Ca2+, P,E2concentration in serum, increase deposition of Ca2+,P in the bone, increase bone density,promote the secretion of E2in vivo, improve the anti osteoporosis related factors in vivo,confirms the salamander general flavone in vivo osteoporosis model mice treated withOPGmRNA, RANKLmRNA, RANKmRNApositive effect.3The experiment proved that the total flavonoids of salamander from molecular biologycurative effect on OVX was compared with other drugs, including dose intervention in OVX,namely: human and animal body surface area equivalent dose, is the ideal dosage, andprovides a theoretical basis for the clinical application of traditional Chinese medicine.
