Objective:This study with6-hydroxydopamine (6-OHDA) stereotactic injection intorat substantia nigra copy model of Parkinson disease, observed Xifengdingchan Pillon the behavior of PD model rats learn; determination Xifengdingchan Pill PD modelrat striatal glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) content; thus Xifengdingchan pill againstoxidative stress injury mechanism study provides a theoretical basis for Chinesemedicine treatment of PD.Methods:Male SD rats30were randomly divided into2groups, stereotactic injectionof6-OHDA into the rat substantia nigra production model of Parkinson disease, thefirst group for the two-point injection group; second group that injected group,relatively little success rate of injection and two injections law making model. Basedon modeling the success rate of the level, select the appropriate method to preparehemiparkinsonism rat model, then packet gavage treatment, divided into five groups:sham operation group,the Xifengdingchan Pill group,Madopar group,XifengdingchanPill+Madopar group,model control group, the observed changes in the behavior ofrats during treatment, involuntary movements (AIM) score. The rats in the lastrotation behavior test after the rats were killed quickly separated to remove thestriatum, the preparation of tissue homogenates using chemical colorimetric detectionof GSH and GSH-Px, SOD, MDA content of the indicators.Results:1. The model of the two-point injection group high success rate at one pointinjection group (P<0.05).2. Two weeks after modeling apomorphine-induced ratmodel of rotation, the number of rotations of rats of the model group, TCM group theMadopar group and combination group were more than7times/minute (Parkinson’sdisease with the internationally accepted standards mouse model). Combination grouptherapy PD rats produce stable contralateral rotation behavior to reduce the occurrence of AIM. Western medicine group after treatment, the PD rats producesunstable contralateral rotation behavior reached a peak of three weeks, and PD rats inapomorphine-induced contralateral rotation behavior gradually increase. Reduction inthe number of joint group allows apomorphine-induced contralateral rotation. Chinesemedicine group and combined group damaged striatum GSH, SOD, GSH-Px contentcompared with the control group increased significantly (P<0.05), and MDA contentwere decreased (P<0.05); alone Madopar treatment GSH, SOD, GSH-Px the contentof MDA and the model group, no significant difference (P>0.05).Conclusion:This study prompts:6-OHDA, through the two-point injection injectionin the rat brain damage striatal system, dopamine content was reduced to simulate theclinical and pathological phenomena, made of Parkinson’s disease model. Has beenshown to have a reliable and reproducible, and higher rate into the mold, and also inline with the pathogenesis and clinical features of PD model. Xifeng Dingchan Pillcombined with Madopar treatment of PD rat model to some extent to reduce theoccurrence of involuntary movements, reducing the amount of Madopar alleviate theside effects of Madopar, at the same time can be significantly reduced PD the ratnigrostriatal oxidative stress injury, its mechanism may have with the side protectionof nigrostriatal antioxidant capacity and free radical scavenging effects.
