Experimented Study Of The Effect Of BushenHuoxueTonglin On Bcl-2and Bax Of Rats Model With BPH

Objective：This project is aimed at the prostate from the index of prostate, apoptosissuppressor gene (Bcl-2) and apoptosis promoting gene (Bax) to study the treatment ofbenign prostatic hyperplasia（BPH）mechanism of Bushen Huoxue Tonglin.To studythe effect of benign prostatic hyperplasia in a rat model of Bcl-2and Bax,and toprovide the scientific theory and experimental basis of benign prostatic hyperplasia.Methods：Male Wistar rats were randomly assigned to sham operation group, modelgroup、positive control group、low-dose group、middle-dose group and high-dosegroup. The Rats were feeding for one week after the start of molding.All groups wereinjectied by10%chloral hydrate (3.5mg/kg), with removal of bilateral intra-abdominaltestes. The operation is asepsis.The sham operation group were free from the removalof the testes.7days later, the sham operation group were injected with10ml/kgphysiological saline,the rest five groups were injected with5mg/kg testosteronepropionate. After modeling, model group were fed with10ml/kg physiological saline,Baoliezhi Group were fed with0.8mg/kg, low-dose group were fed with5ml/kg,middle-dose group were fed with10ml/kg and high-dose group were fed with15ml/kg.All the rats were fed30days,once everyday, Decapitated animals were killed24h afterthe last dose, and take the prostate tissue, packet marking, weighing by electro-opticalanalytical balance, obtaining prostate wet weight. while making light microscopy,immunohistochemistry samples, the specimens of each group were analyzed andobserved prostate cell shape changes under a light microscope.Results：The prostate weight, prostatic index, and Bcl-2expression of3treatment ofrats were significantly lower than the model group,and the expression level of Baxwere significantly higher than model group,particularly in the high-dose group ofBushen Huoxue Tonglin.And compared with positive control group, the the high-doseand middle-dose group of Bushen Huoxue Tonglin had statistics significance, the low-dose group and the positive control group were the Bcl-2expression level hasstatistics significance, the Bax expression level non-statistics significance.The pathological changes of model group was obviously, the pathologicalchanges in the low-dose group and the positive control group were reducer than themodel group. and the high-dose group were most obviously. The histopathologicalchanges in each group has statistics significance.
