【Objective】To investigate the expression of AEG-1in human cercivalcancer and analyze the relationship of AEG-1and clinical pathologicalcharacteristics.【Methods】The expression of AEG-1was examined byimmunohistochemistry(IHC) in normal cercival tissues of21cases, cervicalintraepithelial neoplasia tissues of11cases, cercival cancer tissues of70casesthat consist of squamous carcinoma of58cases and cervicaladenocarcinoma of12cases.【Relsults】A positive stain of Astrocyte elevated gene-1was observed in thecytoplasm and around the nucleus. The immunohistochemical staining shewthat the expression of AEG-1in cervical intraepithelial neoplasia tissues andcercival cancer tissues was higher than that in normal cercival tissues (P<0.05).The expression of AEG-1was associated with the FIGO stage, histologic type,pathologic grade, myometrial invasion and pelvic lymph nodes metastasis, butnot age and histologic type.(P>0.05).【Conclusion】The expression of AEG-1is closely related with the arisingand development of cervical cancer.AEG-1may be an important prognosisfactors of cercival cancer. 【Objective】To investigate the effect and mechanism of siRNA-induced astrocyte elevatedgene-1(AEG-1) downregulation on the malignant phenotype of cervical cancer cell lines.【Methods】An siRNA targeting to AEG-1mRNA was constructed and transfected intocervical cancer cell lines-Hela,Siha and C33a. Cervical cancer cells transfected thenon-specific siRNA were used as negative control group, and untreated cells as blankcontrol group. Reverse transcriptase polymerase chain reaction and Western blot assayswere used to detect the AEG-1,β-catenin,N-cadherin,vimentin and E-cadherin mRNAand protein levels, respectively. The effects of blocking AEG-1on cell proliferation,apoptotic rate were detected by MTT analysis, flow cytometry, and transwell migrationassay, respectively.【Results】The AEG-1mRNA and protein levels both markedly decreased through siRNAas compared with Controls in cervical cancer cell lines-Hela、Siha、C33a. Thedownregulation of AEG-1in Hela cell line was accompanied with epithelial markersdecreased and mesenchymal markers increased.AEG-1siRNA noticeably decreased the cellproliferation and invasion. The apoptosis rate of cervical cancer cell lines transfected withAEG-1siRNA was significantly increased compared with Controls.【Conclusion】AEG-1blockage markedly inhibits cell proliferation and invasion, whilesignificantly promote cell apoptosis. AEG-1regulating the malignant biological behaviorsof cervical cancer cell line may through epithelial-mesenchymal transition (EMT).Hence,AEG-1may be a new target for cervical cancer gene therapy.
