Objective:To investigate the expression of MCM3, Ki67and PCNA and its clinicalsignificance in NSCLC.Method:MCM3, Ki67and PCNA were detected by IHC method in51patients with NSCLC andthe expression of21cases of normal lung tissue as a control. To Calculate threeprotein-positive cell labeling index (Li) in NSCLC, and by Imagepro plus6.0calculating theIOD value of the positive staining region of the three proteins in lung cancer and normallung tissue. Analysis the differences of three proteins labeling index in NSCLC. Statisticalanalysis was carried out by comparing the difference of IOD value of MCM3, Ki67andPCNA in NSCLC and normal tissue, and analysis the association of IOD value andnon-small cell lung cancer clinical and pathological features.Result:1.NSCLC cells were marked in different levels by MCM3, Ki67and PCNA, thedifference of the labeling index of three proteins in NSCLC were significant, and theLabeling index of MCM3were higher than that of Ki67and PCNA.2..MCM3, Ki67and PCNA were expressed in NSCLC with different levels, the positivesignals were located in the nucleus, and the expression of the three proteins in NSCLC wassignificantly higher than in normal lung tissue. 3.The differences of expression of mcm3were significantly among grade1,grade2andgrade3-4NSCLC。The difference of the expression of ki67were significantly betweengrade3-4and grade1NSCLC and between grade2and grade3-4NSCLC,but the differencewas not significant between grade1and grade2NSCLC. The difference of expression ofPCNA were significantly between grade3-4and grade1NSCLC,but the difference werenot significant between grade2and grade3-4NSCLC and grade2and grade3-4NSCLC.The expression of mcm3in large cell lung cancer was significantly higher than that insquamous cell carcinoma (p=0.013) and adenocarcinoma (p=0.037),but the difference ofthe expression of mcm3between large cell carcinoma and adenosquamous carcinoma wasnot significantly,the difference of the expression of mcm3among squamous cell carcinoma,adenocarcinoma and adenosquamous carcinoma was not significantly. The expression ofPCNA in large cell lung cancer was significantly higher than squamous cell carcinoma(p=0.041), but there was no difference among the expression of PCNA in large cell lungcancer, adenocarcinoma and adenosquamous carcinoma, and the difference of theexpression of PCNA among squamous cell carcinoma, adenocarcinoma andadenosquamous carcinoma was not significantly. The difference of the expression of Ki67was not significantly between the different pathological types. The expressions of the threeproteins were not associated with gender, age, tumor size, and lymph node metastasis andtumor pathological stage in NSCLC.4.The expression of MCM3and Ki67were positive correlation with grades ofdifferentiation in NSCLC, the expression of mcm3was more relevant with the grade ofdifferentiated than Ki67in NSCLC. The three proteins expressed in smokers wassignificantly higher than non-smokers in NSCLC.Conclusion1.NSCLC cells were labeled efficiently by MCM3、Ki67and PCNA. The efficiency ofearly diagnosis of NSCLC would be improved if the three proteins were applied in clinicaltesting. 2.Compared to the ki67and PCNA, the expression of MCM3could assess the degree ofdifferentiation, the malignancy and proliferative activity of NSCLC more accurately, it hasa guiding significance for the adjuvant therapy of the postoperative in NSCLC.
